29种中药提取物对耐甲氧西林金黄色葡萄球菌的体外抑菌活性Study on Antibacterial Activity of 29 Traditional Chinese Medicine Extracts against Methicillin-Resistant Staphylococcus Aureu

摘要(Abstract):
目的:评估29种中药水提物和醇提物对耐甲氧西林金黄色葡萄球菌(MRSA)的体外抑菌活性,筛选抑菌活性最高的中药做进一步研究。方法:用K-B纸片法筛选出有抑菌活性的中药;用微量稀释法测定中药的最低抑菌浓度(MIC)和最低杀菌浓度(MBC);并用微量棋盘稀释法测定联合用药对MRSA的抑制作用。结果:K-B纸片法筛选结果显示,29种中药水提物仅黄柏有较小的抑菌圈,7种中药的醇提物有抑菌圈,其中黄柏的抑菌圈直径最大,均值为18.1 mm;金银花和紫花地丁抑菌圈不透明; 29种中药醇提物黄柏对MRSA的抑菌活性最强,其MIC的生药浓度为125 g/L,MBC的生药浓度为250 g/L;甘草与黄苓联合应用时,抗菌指数FIC <0.5。结论:所选中药醇提物对MRSA的抑菌活性强于水提物,黄柏醇提物最能有效抑制MRSA的生长;甘草与黄苓联合应用具有协同作用,对MRSA的抑菌活性最高。
Objective: To evaluate antibacterial activity of traditional Chinese medicine water extracts and ethanol extracts against methicillin-resistant staphylococcus aureu( MRSA) in vitro,screening the traditional Chinese medicine with the highest antibacterial activity for further study. Methods: The Chinese medicine with the powerful antibacterial activity against MRSA were selected by K-B paper dispersion; The minimal inhibitory concentration( MIC) and minimal bactericidal concentration( MBC) were determined by micro dilution method; The microdilution and checkerboard method were used to determine inhibitory effect of the Chinese medicine combination against MRSA. Results: Only water extracts of Golden Cypress had the smaller inhibition zone,ethanol extracts of 7 traditional Chinese medicine had inhibition zone, and the Golden Cypress had the biggest and transparent inhibition zone,18. 1 mm; Honeysuckle and Herba violae had the non-transparent inhibition zone. The results showed that the Golden Cypress ethanol extracts had the strongest antibacterial activity,the crude drug concentration of MIC was 125 g/L; that of MBC was 250 g/L. When Liquorice was combined with Radix Scutellariae,FIC index was < 0. 5. Conclusion: The ethanol extracts have stronger antibacterial activity than the water extracts to MRSA in vitro. The MIC and MBC of Golden Cypress ethanol extracts have minimum and thereby inhibit the growth of MRSA most effectively.According to the judgment standard of the FIC index,the antibacterial activity of Liquorice combined with Radix Scutellariae in vitro performances as synergistic effect against MRSA,and the antibacterial activity is the highest.
