不同剂量链脲佐菌素建立1型糖尿病大鼠模型
A Study on the Establishment of Type 1 Diabetes Rat Models with Different Doses of Streptozotocin

摘要(Abstract):

目的:观察不同剂量链脲佐菌素(STZ)致SD大鼠1型糖尿病(T1DM)模型状况的差异,探讨STZ建模的最佳剂量。方法:80只雄性SD大鼠随机分为4组,3个实验组分别一次性尾静脉注射40、50、60mg/kg的STZ,对照组注射等量的枸橼酸盐缓冲液;监测各组大鼠空腹血糖、尿糖、饮水量、采食量、排尿量和体重等指标。结果:D40组(40mg/kgSTZ组)、D50组(50mg/kgSTZ组)和D60组(60mg/kgSTZ组)的成模率分别为70.0%、100.0%、100.0%;死亡率分别为:0.0%、10.0%、60.0%。各成模大鼠均有"三多一少"的糖尿病临床表现。D40组大鼠空腹血糖和尿糖水平2周后有明显下降趋势,而D50组及D60组大鼠一直维持在较高水平。结论:尾静脉注射50mg/kgSTZ成模率高、死亡率低、模型稳定,是建立T1DMSD大鼠模型的最佳剂量。
Objective:To compare the differences of type 1 diabetes mellitus(T1DM)SD rat models induced by different doses of streptozotocin(STZ)and to optimize the dose for establishment of T1DM models.Methods:Eighty male SD rats were randomly divided into 4 groups including 3 diabetic groups which were injected with 40,50,and 60 mg/kg of STZ(groups D40,D50,and D60)once through caudal vein respectively,and 1 control group which was injected with citrate buffer solution of the same volume.The fasting blood glucose,urine glucose,and daily water intake,urinary output,and food intake,and body weight of rats in all the groups were observed.Results:The model success rates of groups D40,D50,and D60 were 70.0%,100.0%,and 100.0%,and the death rates were 0.0%,10.0%,and 60.0% respectively.All of the rats in the 3 groups showed clinical manifestation of diabetes mellitus.The fasting blood glucose and urine glucose levels had downtrend in group D40 in 2 weeks,but kept high level in groups D50 and D60.Conclusion:The dose of 50 mg/kg of STZ is the best amount for establishment of T1DM rat models by injection through caudal vein which leads to higher success rate,lower death rate,and stable clinical features.
