口腔鳞癌中肿瘤相关巨噬细胞、血管内皮生长因子与血管生成的相互关系(英文)
The Relationship among Tumor-associated Macrophages,VEGF and Angiogenesis in Oral Squamous-cell Carcinomas

摘要(Abstract):

目的:研究人口腔鳞癌中肿瘤相关巨噬细胞(tumor-associated macrophages,TAMs)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在血管生成中的关系。方法:收集34例人口腔鳞癌组织标本,光学显微镜进行巨噬细胞和微血管的计数,图像分析软件检测VEGF的表达;8例正常口腔黏膜为对照组。结果:在口腔鳞癌中,VEGF的表达和微血管计数、巨噬细胞计数呈现正相关(P<0.05);VEGF的表达、巨噬细胞的浸润和肿瘤的淋巴结转移有关(P<0.05),口腔癌浸润的巨噬细胞中有29%~10%的细胞内有VEGF的表达。结论:VEGF的表达、巨噬细胞的浸润和血管生成之间具有相关性,二者共同参与了肿瘤的生长和转移。
Objective:To study the correlations among tumor-associated macrophages (TAMs),vascular endothelial growth factor (VEGF) and angiogenesis in human oral squamous-cell carcinomas (OSCC).Methods:Tissue specimens of 34 cases of human oral squamous-cell carcinoma (OSCC) were stained with a standard labelled avidin-biotin (SP) technique and macrophages and microvessels were counted under light microscope after the staining. The expression of VEGF was observed by using image analysis and the secretion of VEGF by macrophagus was tested by using immunohistochemistry method. Eight specimens of healthy oral mucosa (control group) were treated with the same methods. Data were analyzed statistically. Results:In OSCC,VEGF expression positively correlated with counts of microvessels and macrophages(P<0.05),and VEGF expression and microphage counts significantly associated with lymphatic metastasis(P<0.05). From 29.0% to 10.0% of TAMs in OSCC secreted VEGF. Conclusions:The relationship among VEGF expression,macrophage infiltration,and angiogenesis indicates an important synergistic inter-relation between tumor cells and host cells in the micro-environment of OSCC. They both might be involved in the growth and metastasis of the tumor.
