应用PcAb—ABC—Dot—ELISA检测弓形虫CAg的研究The Detection of Toxoplasma Gondii Circulating Antigen by PcAb-ABC-Dot-ELISA

摘要(Abstract):
检测宿主体内的CAg可以早期诊断寄生虫感染,鉴于应用PcAb-ABC-Dot-ELISA检测弓形虫CAg国内尚未见报道,作者用生物素化抗弓形虫PcAb为探针,检测了15份人工感染弓形虫的兔血清,结果15份血清的CAg均呈阳性反应。用35份正常兔血清,10份自然感染球虫的兔血清及血吸虫、华校睾吸虫感染的兔血清各5份作对照,除一份正常兔血清及一份血吸虫感染的兔血清呈阳性反应外,其余均为阴性。结果表明,该法有较好的特异性及重复性。
Although the T. gondii circulating antigen was successfully detected with McAb-ELISA, the utilization of PcAb-ABC-Dot-ELISA jn detecting it has not been reported yet. Using biotin labelled PcAb against T. gondii as a probe, an experiment was conducted to detect the circulating antigen in sera of infected rabbits with PcAb-ABC-Dot-ELISA. The results were as follows. All the sera from 15 infected rabbits showed positive reactions on the 13th day after infection whereas only one of the 35 normal controls gave a posititive reaction. 10 sera from the rabbbit Infected with Eimeria sp, 5 with C. sinensis and 5 with S. japunicum weretested. Among them, only one of those infected with S. japonicum show ed a postive reaction. Our studies showed that the PcAb-ABC-Dot-ELISA is a sensitive and specific method and it's reproducibility is good for detecting CAg of T. gondii in the sera.
