不同剂量链脲佐菌素建立妊娠糖尿病大鼠模型的研究A Study on Establishment of Gestational Diabetes Rat Model with Different Doses of Streptozotocin

摘要(Abstract):
目的:用不同剂量链脲佐菌素(STZ)建立妊娠糖尿病大鼠模型,探讨最佳剂量。方法:SD大鼠按照1∶2雄雌比例合笼过夜,阴道涂片镜检见精子者记为妊娠0d(d0),标记孕鼠并计算孕期,将55只孕鼠随机分成3组;孕期第6天(d6)早晨,实验组分别用45mg/kg(D45)和55mg/kg(D55)的STZ一次性腹腔注射,对照组用等量枸橼酸盐缓冲液代替,处理前禁食12h,观察各组大鼠空腹血糖、尿糖、成模率、死亡率及转阴率。结果:与对照组相比,给药后实验组空腹血糖、尿糖均有明显升高。45mg/kg和55mg/kg成模率分别是95%和100%,转阴率分别是10.52%和6.67%,死亡率分别是5%和33.3%,两实验组比较仅死亡率有显著性差异。结论:腹腔注射STZ45mg/kg成模率高,死亡率低,转阴率低,是建立妊娠糖尿病大鼠模型的理想剂量。
Objective:To establish gestational diabetes (GD) rat model with different doses of streptozotocin(STZ),and to investigate the optimal dose. Methods:Female SD rats and male rats were caged together with the proportion of 1:2. The time when sperm cells were observed in female rats under microscope was recorded as pregnancy 0d. Fifty five pregnant rats were randomly divided into 3 groups. On the 6th day of pregnancy after fasting 12 h,GD groups were injected with 45 mg/kg (group D45) or 55mg/kg (group D55) of STZ once through abdominal cavity respectively and control group was injected with citrate buffer solution instead. Fasting blood glucose,urine glucose,success rate,mortality and negative conversion rate were observed. Results:Compared with those of control group,blood glucose levels and urine glucose levels of GD groups increased significantly. Success rates of groups D45 and D55 were 95.0% and 100.0% respectively; Death rates and negative conversion rates were 5.0%,33.3% and 10.52%,6.67% respectively. The only statistically significant difference between group D45 and group D55 was mortality. Conclusion:The ideal dose of STZ for establishment of GD rat model is about 45mg/kg by injection through abdominal cavity,which leads to higher success rate,lower death rate,and lower negative conversion rate.
