贵州天名精全草中乙酸乙酯部位化学成分的分离鉴定
Isolation and identification of chemical constituents of ethyl acetate in the whole herb of Guizhou Carpesium faber

摘要(Abstract):

目的 探讨贵州天名精全草中乙酸乙酯部位化学成分的分离鉴定。方法 贵州天名精全草30 kg,乙醇浸泡提取,乙酸乙酯萃取后得浸膏1.5 kg,采用硅胶柱层析、MCI gel CHP 20P柱层析、葡聚糖凝胶LH-20柱层析、Rp-C18柱层析、制备薄层色谱、制备型以及半制备型高效液相色谱仪(HPLC)等多种现代色谱分离手段对贵州天名精的乙酸乙酯部位化学成分进行分离纯化,采用超高效液相色谱-四极杆-飞行时间串联质谱技术、核磁共振谱仪以及文献对照等方法,对分离所得化合物结构进行鉴定。结果 从贵州天名精全草乙酸乙酯层分离鉴定了9个化合物,分别为3β-羟基-5α,6α-环氧-7-大柱香波龙烯-9-酮(1)、8,10-二羟基-9-异丁酰基百里香酚(2)、8-羟基-9,10-二异丁酰氧基麝香草酚(3)、毛脉五味子醇(4)、(9Z,12S,13E,15Z)-12-Hydroxyoctadeca-9,13,15-trienoic-acid-methylester(5)、植醇(6)、5-hydroxy-5,6-secocaryophyllen-6-on(7)、5′-甲氧基松脂素(8)、丁香树脂素(9)。结论 从贵州天名精全草乙酸乙酯层分离鉴定了9个化合物,化合物1、4、5、7及9为首次从贵州天名精中分离得到。
Objective To investigate the isolation and identification of the chemical constituents of ethyl acetate from the whole herb of Guizhou Carpesium faber. Methods A total of 30 kg of the whole herb of Guizhou Carpesium faber was extracted by ethanol immersion, and 1.5 kg extractum was extracted with ethyl acetate. The chemical components of the ethyl acetate of Guizhou Carpesium faber were separated and purified by silica gel column chromatography, MCI gel CHP 20P column chromatography, glucan gel LH-20 column chromatography, Rp-C18 column chromatography, preparative thin layer chromatography, preparative and semi-preparative high performance liquid chromatography(HPLC) and other modern chromatographic separation methods, and the structure of the isolated compounds was identified by UHPLC-Q-TOF-MS/MS, nuclear magnetic resonance spectrometry and literature reference. Results Nine compounds were isolated and identified from the ethyl acetate layer of the whole herb of Guizhou Carpesium faber, which were 3β-Hydroxy-5α,6α-epoxy-7-megastigmen-9-one(1), 8,10-dihydroxy-9-isobutyrylthymol(2), 8-hydroxy-9,10-diisobutyryloxythymol(3), PubinernoidA(4),(9Z,12S,13E,15Z)-12-hydroxyoctadeca-9,13,15-trienoic-acid-methylester(5),Phytol(6),5-hydroxy-5,6-secocaryophyllen-6-on(7), 5′-methoxyrosinol(8), syringaresinol(9). Conclusion Nine compounds were isolated and identified from the ethyl acetate layer of Guizhou Carpesium faber, of which compounds 1, 4, 5, 7, and 9 were isolated for the first time from Guizhou Carpesium faber.
