太子参对急性心肌梗死心力衰竭大鼠心肌重构的作用
Effects of Radix Pseudostellariae on Cardiac Remodeling in Rats with Chronic Heart Failure Induced by Acute Cardiac Infarction and Its Mechanism

摘要(Abstract):

目的:研究太子参对左冠状动脉前降支结扎所致急性心肌梗死慢性心衰大鼠心肌重构的作用,并探讨其可能的机制。方法:设假手术组、模型组、太子参高剂量组和低剂量组,后3组结扎大鼠冠状动脉导致急性心肌梗死(AMI)复制慢性心衰大鼠动物模型,造模后1周,太子参组开始给药5周;以死亡率、心肌重构指数分析心肌重构的程度,酶联免疫吸附试验(ELISA)分析血清中IL-6和TNF-α的水平。结果:冠脉结扎6周后,模型组大鼠死亡率高达43.8%,心肺指数、心肌重构指数及血清中IL-6和TNF-α的水平显著增加,与假手术组比较,差异显著;而太子参组上述指标具有显著改善作用。结论:太子参对急性心肌梗死诱发的慢性心衰具有一定的保护作用,其作用可能与太子参影响细胞因子有关。
Objective:To investigate the effects of Radix Pseudostellariae on cardiac remodeling of chronic heart failure induced by acute cardiac infarction after coronary artery ligation and its mechanism. Methods:Rat chronic heart failure was produced by coronary artery ligation of 6 weeks. The cardiac remodeling was evaluated with remodeling indexes such as left and right ventricular indexes,lung index,and globular index,etc. The death ratio of model rats was used to indicate the heart failure degree. Cytokine of IL-2 and TNF-α were detected by ELISA to explore the mechanism of Radix Pseudostellariae. Results:During the process of heart failure caused by coronary artery ligation and the heart muscle remodeling,the death rate of model rats was 43.8%,heart and lung indexes,heart remodeling index and serum levels of IL-6 and TNF-α were significantly higher in modeling group than those of sham operation group. After the treatment of Radix Pseudostellariae decoction by ig for continuous 5 weeks,the death rate,cardiac remodeling indexes,and serum levels of IL-6 and TNF-α markedly decreased. Conclusion:Long-term use of Radix Pseudostellariae can ameliorate cardiac remodeling of the chronic heart failure induced by acute cardiac infarction and the one of the mechanisms might be its influence to cytokine.
