三氧化二砷对大鼠血细胞参数影响的实验分析

摘要(Abstract):

目的:分析三氧化二砷(AS2O3)对实验大鼠血细胞15项参数的影响,为研究砷对血液系统的影响及其作用机制提供参考依据。方法:AS2O3经口染毒,建立砷中毒动物模型,采用F-820半自动血细胞分析仪测定实验动物血细胞参数的变化。结果:(1)红细胞参数:红细胞计数(RBC)、血红蛋白(HGB)、红细胞压积(HCT)显著降低,平均红细胞血红蛋白含量(MCH)显著升高,差异具有统计学意义(P<0.05);平均红细胞血红蛋白浓度(MCHC)随剂量增加而升高,其中高剂量组差异具有统计学意义(P<0.05);(2)白细胞参数:低、中剂量组白细胞计数(WBC)显著增高,差异具有统计学意义(P<0.05),但随染砷剂量的增加而呈下降趋势,中剂量组淋巴细胞百分数(W-SCR)显著降低,差异具有统计学意义(P<0.01);中性粒细胞百分数(W-LCR)出现低、中剂量组随剂量增加而显著升高,差异具有统计学意义(P<0.01),但高剂量组有降低的现象;淋巴细胞绝对值(W-SCC)及中性粒细胞绝对值(W-LCC)出现低剂量显著升高而随染砷剂量的增加而逐渐降低的趋势;(3)血小板参数:各AS2O3组血小板(PLT)呈下降趋势,其中以中、高剂量组下降显著,差异具有统计学意义(P<0.05);而血小板分布宽度(PDW)、平均血小板体积(MPV)指标呈升高趋势,其中低、中剂量组显著升高,差异具有统计学意义(P<0.05)。结论:血液系统可能是砷毒性作用较为敏感的靶系统,AS2O3可对实验大鼠血液系统产生一定的损害作用,引起大鼠血液系统红细胞、白细胞、血小板等指标发生改变。
Objective: To anslyze changes of 15 blood parmeters under the influence of arsenic trioxide(AS2O3) in experimental rats,so as to provide references for further study.Method: Arsenic poisoning animal model was established by given AS2O3 to rats orally.F-820 semi-automatic blood corpuscle analyzer was used to anslyze blood parameters of the experimental animals.Results: Compared with normal control group,some changes of the checked blood parameters were found in arsenic poisoning groups:(1)RBC,HGB,and HCT reduced and MCH increased significantly(P<0.05 or P<0.01).MCHC increased with the rising of drug doses used.The difference was significant for the high dose group(P<0.05).(2)WBC of groups received drug doses of 1.25 mg/kg and 2.5 mg/kg increased obviously,and the differences were significant(P<0.05 or P<0.01).But when doses increased more,it descended,and in 5 mg/kg group it was about the level of normal control group.W-SCR reduced significantly in the 2.5 mg/kg group(P<0.01).W-LCR increased in low and middle dose groups(P<0.01),but decreased in the 5 mg/kg group.W-SCC and W-LCC increased in low dose group,but when dose levels rose more,W-SCC and W-LCC decreased;(3)PLT showed a declining trend in each group.The differences were significant for 2.5 mg/kg and 5 mg/kg groups(P<0.05).PDW and MPV showed a rising trend,specially in 1.25 mg/kg and 2.5 mg/kg groups(P<0.05).Conclusions: Blood system may be a sensitive target system to the influence of arsenic toxic.AS2O3 has harmful effect on blood system of experimental rats to some extent,and can cause changes of indexes including hematid,leucocyte and platelet,but the mechanisms need to be studied further.
