从保存血液中分离淋巴细胞的某些条件观察

摘要(Abstract):

保存后的血液,经淋巴细胞分离液密度梯度离心分离所获得的单形核细胞中,粒细胞和红细胞污染严重。室温保存48h 血液分离所得淋巴细胞,较之4℃保存所分离的粒细胞污染较轻,而红细胞污染较重。降低淋巴细胞分离液密度,有助于改善粒细胞和红细胞的污染,但同时伴有细胞收获量的减少。室温保存48h 血液,其淋巴细胞 HLA 抗原特异性的表达和新鲜淋巴细胞一致。
Mononuclear cells separated from stored blood were highly contaminated with polynuclears and erythrocytes by density gradient separation.The lymphoeytes separated from blood stored at 4℃ were better at erythrocyte but worse for polynuclear cells contaminations than those at room temperature.Diminishing the density of lymphocyte separation medium could slightly improve the qua- lity but decrease the yield of lymphocyte simultaneously. Lymphocytes separated from blood stored at room temperature 48 hours expressed their surface HLA antigens as perfect as the fresh ones.
