良附滴丸抗大鼠乙醇型胃溃疡预防作用及机制

摘要(Abstract):

目的:研究良附滴丸对乙醇型胃溃疡大鼠模型的预防作用及机制。方法:48只SD大鼠均分为良附滴丸(低、中、高)剂量组、奥美拉唑组、模型组以及空白对照组,低、中、高剂量组分别灌胃良附滴丸[良附滴丸剂量0. 27 g/(kg·d)、0. 54 g/(kg·d)、1. 08 g/(kg·d)],奥美拉唑组灌胃奥美拉唑[18 mg/(kg·d)],模型组和空白组给予等体积生理盐水,连续给药5 d,每天给药1次,第5天给药后禁食不禁水24 h,再给药1次,末次给药1 h后,除空白对照组外,其余各组大鼠灌胃无水乙醇(5 m L/kg)诱导胃溃疡,于1 h后处死大鼠并收集胃组织,观察各组大鼠胃组织表观形态和HE染色下组织形态特征,比较胃组织溃疡指数、计算溃疡抑制百分率,检测胃组织匀浆中一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、前列腺素(PGE2)等指标的含量。结果:与模型组比较,良附滴丸中、高剂量组可改善胃溃疡组织水肿、黏膜糜烂、血管扩张出血及炎症细胞浸润等病理改变;良附滴丸低、中、高剂量组的溃疡抑制百分率依次升高;与模型组比较,良附滴丸低剂量组能够升高NO、SOD的含量(P <0. 05或P <0. 01),降低IL-6、TNF-α含量(P <0. 05或P <0. 01),对MDA、PGE2的含量影响不明显(P> 0. 05);良附滴丸中、高剂量组能降低胃黏膜损伤大鼠胃溃疡指数(P <0. 01),胃组织中NO、SOD、PGE2含量明显升高(P <0. 01),TNF-α、IL-6、MDA水平明显降低(P <0. 05或P <0. 01)。结论:良附滴丸可减轻大鼠无水乙醇诱导的胃溃疡病理改变,其机制可能与增加NO、SOD、PGE2水平或降低TNF-α、IL-6、MDA水平有关。
Objective: To investigate the preventive effect and its mechanism of liangfu dripping pills on ethanol-induced rat gastric ulcer. Methods: Based on the treatment reagents,48 SD rats were randomly divided into liangfu dripping pills low [0. 27 g/( kg·d) ],medium [0. 54 g/( kg·d) ]and high [1. 08 g/( kg · d) ] dose groups,omeprazole group [18 mg/( kg · d) ],the model group( absolute ethanol) and the blank group( administer equal volume of normal saline). Rats were treated once a day for 5 continuous days. After the treatment on 5 thday,rats were given water only for 24 h and then given the last administration. At 1 h after the last administration,except for the blank group,rats were intragastrically administrated with absolute ethanol( 5 m L/kg) to induce gastric ulcers for1 h. The rats were sacrificed and gastric tissues were collected to observe gastric tissue morphology.H&E staining was used to detect morphological characteristics for comparing gastric tissue ulcer index and calculating inhibition percentage of ulcers. Nitric oxide( NO), malondialdehyde( MDA),superoxide dismutase( SOD), tumor necrosis factor-α( TNF-α), interleukin-6( IL-6), and prostaglandin( PGE2) in gastric tissue homogenate were measured. Results: Compared with the model group,the medium-and high dose groups of liangfu dripping pills reduced the pathological changes including gastric ulcer tissue edema,mucosal erosion,vasodilation and inflammatory cell infiltration.Liangfu dripping pills inhibited ulcers in dose-dependent manner. Liangfu dripping pills upregulated NO and SOD( P < 0. 05 or P < 0. 01) and reduced IL-6 and TNF-α( P < 0. 05 or P < 0. 01).Medium-and high doses of liangfu dripping pills decreased the gastric ulcer index( P < 0. 01),upregulated NO,SOD and PGE2( P < 0. 01),while downregulated TNF-α,IL-6 and MDA( P < 0. 05 or P < 0. 01). Conclusion: Liangfu dripping pills alleviate the pathological changes of rat gastric ulcer induced by absolute ethanol. The mechanism may be related to upregulation of NO,SOD and PGE2 or downregulation of TNF-α,IL-6 and MDA.
