能量合剂对脑血栓急性形成期大鼠脑神经细胞的保护作用

摘要(Abstract):

目的:探讨能量合剂对脑血栓急性形成期大鼠脑神经细胞的保护作用。方法:健康SD大鼠200只,随机选取160只分为治疗组和模型组,另40只作为对照组,治疗组和模型组使用三氯化铁法制备大鼠脑血栓模型,对照组为假手术组;造模后治疗组大鼠肌肉注射能量合剂,模型组及对照组大鼠注入等量的生理盐水;于造模后3、6、12、24 h分别处死大鼠,用红四氮唑(TTC)染色法和流式细胞术检测大鼠脑梗死面积和脑细胞凋亡情况。结果:造模后6、12、24 h治疗组大鼠脑梗死面积小于模型组,凋亡细胞百分率小于与模型组,差异具有统计学意义(P<0.05)。结论:脑血栓急性形成期使用能量合剂可减少脑组织梗死面积,抑制脑细胞凋亡。
Objective: To investigate the protective effects of energy mixture on brain cells of rats at acute cerebral thrombosis formation stage. Methods: A total of 160 SD rats were selected and randomly divided into treatment group and model group( 80 in each),and another 40 SD rats severed as control group. Ferric chloride methods was used to establish cerebral thrombosis model in treatment group and model group,and control group was sham operation group. After modeling,treatment group was given energy mixture,model and control groups were given normal saline instead. In 3,6,12,24 h after modeling,rats were sacrificed,infarction area and apoptosis of brain cells were detected by using triphenyltetrazolium chloride( TTC) staining and flow cytometry respectively. Results: In 6,12,24 h after modeling,the infarction areas in treatment group were bigger than those of model group,and apoptosis cell ratios in treatment group were lower than those of model group. The differences were statistically significant( P < 0. 05). Conclusions: Using energy mixture in acute cerebral thrombosis formation stage can reduce brain infarction area,inhibit brain cell apoptosis,and has a protective effect on survival brain cells.
