摘要(Abstract):
目的:动态观察纤溶酶原激活物抑制剂-1(PAI-1)和金属蛋白酶组织抑制因子-1(TIMP-1)在糖尿病大鼠肾脏中的表达,探讨PAI-1及TIMP-1在糖尿病肾病(DN)肾纤维化发生、发展过程中的作用。方法:链脲菌素(STZ)诱导大鼠糖尿病肾病,免疫组织化学的方法动态观察糖尿病肾病发生、发展过程中大鼠肾脏PAI-1、TIMP-1及纤维连接蛋白(FN)表达。结果:PAI-1、TIMP-1及FN表达从糖尿病2周起较正常对照组明显增多(P<0.01),PAI-1和TIMP-1阳性表达主要位于肾小管上皮细胞胞浆内,FN阳性表达主要位于肾小管间质和肾小球,且随着糖尿病进展有逐渐增加的趋势。结论:PAI-1和TIMP-1在肾小管上皮细胞的高表达可能参与了糖尿病肾病过程中肾纤维化的形成。
Objective: To observe the expression changes of plasminogen activator inhibitor-1(PAI-1) and tissue inhibitor of metalloproteinase-1(TIMP-1) in kidney of diabetic nephropathy rats,and to explore the role of PAI-1 and TIMP-1 in the development of renal fibrosis of diabetic nephropathy(DP).Methods: DP in rats was induced by tail intravenous injection of streptozocin(STZ).The expression of PAI-1,TIMP-1 and fibronection(FN) protein were examined with immunohistochemical technique.Results: Expression of PAI-1,TIMP-1 and FN in kidney of diabetic rats began to increase in the second week of diabetes;PAI-1 and TIMP-1mainly expressed in intracytoplasm of renal tubular epithelial cells,while FN mainly expressed in renal tubule matrix and glomerului;Their expression showed a tendency of increasing along with the progression of diabetes.Conclusions: The high expression of PAI-1 and TIMP-1 in renal tubular epithelial cells might be involved in renal fibrosis of DP.
