摘要(Abstract):
目的:观察10Hz,15Hz,25Hz,50Hz频率的阈下电刺激对缺血心肌血管新生的影响,寻找产生缺血心肌血管新生的有效阈下电刺激频率。方法:将30只心肌缺血模型大鼠随机分为阈下电刺激频率10Hz,15Hz,25Hz,50Hz组与对照组,用免疫组织化学法检测缺血心肌中毛细血管内皮细胞数、毛细血管密度。结果:(1)与对照组比较,25Hz组、50Hz组大鼠缺血心肌中毛细血管内皮细胞数、毛细血管密度显著增加(P<0·01);(2)与对照组比较,10Hz组、15Hz组大鼠缺血心肌中毛细血管内皮细胞数、毛细血管密度差异无显著性。结论:25Hz,50Hz频率的阈下电刺激可促进大鼠缺血心肌中毛细血管的新生。
Objective:To study the effect of electrical stimulation below contraction threshold to angiogenesis in ischemic myocardium of rat.Methods:Myocardial infarct model was experimentally established with 30 wistar rats.Then,these rats were randomly divided into control group(did not receive electrical stimulation),and electrical stimulation groups,which were implanted with electrodes and received stimuli with frequencies of 10 Hz,15 Hz,25 Hz,and 50 Hz respectively.The stimulus strength was 0.3 V,which was far below the threshold of myocardial contraction.Endothelial cell(EC)and capillary density(CD)were detected with immunoistochemical method.Results:The numbers of EC and CD of ischemic myocardium in 25 Hz group and 50 Hz group were significantly higher than those in control group(P<0.01);while the numbers of EC and CD of ischemic myocardium in 10 Hz group and 15 Hz group did not significantly differ from those of control group.Conclusions:Electrical stimulation below contraction threshold at frequencies of 25 Hz and 50 Hz may promote angiogenesis in ischemic myocardium of rats.
