抗人红细胞单克隆抗体的制备和鉴定

摘要(Abstract):

用O型人红细胞免疫的Balb/c小鼠脾细胞和NS--1骨髓瘤细胞融合、克隆后,获得两株杂交瘤细胞株。两株细胞分泌的单克隆抗体都是IgG_s亚类。文中对此类抗体的特性进行探讨。吸收试验表明,这两种单克隆抗体并不是和红细胞H抗原反应,但可能是同ABO各型红细胞上一种共同抗原反应。
Two strains of hybridoma secreting McAb against human RBC were obtain- ed by fusing meyloma cells NS-1 with splenocytes from Balb/c mice imm- unized by human group O erythrocytes. The monoclonal antibodies secreted by these cells only reacted with human erythrocytes of any ABO homotype, but not with the erythrocytes of animals, including monkey, cat, dog, pig etc. Their hemoagglutination titers was 1/32-1/128 for culture supernatants, H4E10- S &H4D1-S anp 1/4096-1/32768 for ascites H4D1-A and H4E10-A. They are all IgG3 subclass. H4E10-A can induce the immune hemolysis of ythrocytes when the undiluted rabbits sera are used as the complement but not diluted rabbits and guinea pig sera. Absorption tet revealed that the antiqen human errec ognized by these two McAbs is a common antigen of human erythrocytes bas nott H anigen.
