多聚赖氨酸与EDTA交联物对小鼠成纤维细胞的毒性研究Toxicity Study of Conjugates of PLL and EDTA on Mouse Fibroblast

摘要(Abstract):
目的:观察多聚赖氨酸与乙二胺四乙酸(EDTA)的交联物(PLE)对体外培养的小鼠成纤维细胞L929的毒性程度。方法:小鼠成纤维细胞L929分为正常组、对照组、多聚赖氨酸组、PLE组和EDTA组,采用MTT法比较相同浓度下,EDTA、多聚赖氨酸与EDTA交联物对小鼠成纤维细胞L929相对增殖率的影响。结果:以正常组作为标准(100%),多聚赖氨酸组、EDTA组、PLE组和对照组对小鼠成纤维细胞L929的相对增殖度(RGR)分别为79.87%、69.35%、81.47%和20.12%,均较正常组下降,差异有统计学意义(P<0.05);在质量浓度达为1 000μmo L/L时,EDTA对小鼠成纤维细胞L929的毒性为2级,PLE和多聚赖氨酸对小鼠成纤维细胞L929的毒性为1级。结论:实验浓度的PLE和EDTA对小鼠成纤维细胞L929毒性较低,具有一定的安全性。
Objective: To investigate the toxicity degree of PLE from EDTA and Poly-L-Lysine( PLL) on fibroblast L929 of mouse in vitro culture. Methods: Mouse fibroblast L929 were divided into normal group,control group,PLL group,PLE group and EDTA group,adopting MTT method to compare the effect of EDTA and PLE on proliferation of mouse fibroblast L929 under the same concentration. Results: Taking normal group as standard( 100%),relative growth rate( RGR) of L929 in PLL group,EDTA group,PLE group and control group were 79. 87% 、69. 35% 、81. 47% and20. 12%,lowered than normal group,differences were statistically significant( P < 0. 05). As the mass concentration reached 1 000 μmo L / L,toxicity of EDTA on fibroblast L929 reached level 2,PLE group reached level 1. Conclusions: Experiment concentration PLE and EDTA has minor toxicity on mouse fibroblast L929.
