黄芪当归在肾小管间质纤维化大鼠模型中的作用Effects of A Mixture of Astragalus Membranaceus and Angelica Sinensis on Rats with Renal Tubulo - intersititial Fibrosis

摘要(Abstract):
目的:探讨黄芪、当归在肾间质纤维化大鼠模型中的作用及机制。方法:采用单侧输尿管结扎木(UUO)致肾间质纤维化大鼠模型,将大鼠随机分为4组:假手术组、模型组、依那普利(ACEI)治疗组、黄芪当归治疗组。手术后第9天处死大鼠,经免疫组化方法观察各组大鼠肾脏平滑肌肌球蛋白(α-SMA)的表达,Masson染色评定各组肾小管间质损伤指数。结果:模型组α-SMA的表达及肾小管间质损伤指数明显高于假手术组(P<0.01),而ACEI治疗组、黄芪当归治疗组明显低于模型组(P<0.01),两治疗组间无显著差别(P>0.05)。结论:黄芪、当归合剂可通过抑制成肌纤维细胞的形成,在肾间质纤维化中发挥和ACEI相类似的肾保护作用。
Objective: To investigate the effects of a mixture of Astragalus membranaceus and Angelica sinensis (AA)on rats with renal tubulo - interstitial fibrosis and the mechanism of it. Methods: Rats were randomly divided into 4 groups: sham group ( SH) , model group ( MD ) , ACEI treatment group ( AC) , and As. membranaceus and An. sinensis treatmenttreatment group ( AA ) . Rat models were made with unilateral ureteral obstruction (UUO). 9 days after UUO, the expression of α-SMA in obstructed kidney were measured with immunohistochemistry. At same time, the degree of tubulo - interstitial damage ( DTD) was scored. Results: The level of α-SMA and DTD in group MD increased markedly compared with those of group SH (P <0. 01 ) . These 2 indexes in groups AC and AA decreased in comparison with those of group MD (P <0. 01) . The level of a-SMA was significantly reduced by both ACEI and AA, and there was no difference of level of a-SMA between groups of AC and AA(P >0. 05) . Conclusion:Similar to ACEI, AA may prevent fibrosis development. This might produce partly via preventing die promotion of myofibroblast.
