天葵提取物对小鼠免疫功能的影响

摘要(Abstract):

目的:探讨天葵提取物(TK)对小鼠免疫功能的影响。方法:通过TK急性毒性实验,测定TK对小鼠的半数致死量(LD50);采用碳粒廓清法观察TK对小鼠单核巨噬细胞吞噬功能的影响,并计算肝脏、胸腺和脾脏指数;采用半数溶血值HC50法测定血清溶血素的形成,并计算胸腺指数和脾脏指数;采用环磷酰胺(CTX)复制免疫抑制模型,观察TK对小鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、尿素氮(BUN)的影响,并计算胸腺和脾脏指数。结果:在TK最大给药浓度和最大给药体积下,给药组小鼠生长良好,未见死亡;免疫实验表明,与对照组相比,给药组小鼠巨噬细胞的吞噬指数α、HC50、脾脏指数显著升高,差异具有统计学意义(P<0.05);与CTX诱导的免疫抑制模型组相比,给药组血清中的AST、ALT、BUN含量明显降低(P<0.05),脾脏指数明显升高(P<0.05)。结论:TK能增强小鼠的免疫功能,且对CTX引起的小鼠肝肾功能损伤有一定的保护作用。
Objective: To investigate the impact of Semiaguilegia adoxoides extract( TK) on immune function of mice. Methods: LD50 of TK was determined by acute toxicity testing. The effect of TK on phagocytosis of mononuclear macrophage of mice was determined by carbon particle clearance test and the indexes of liver,thymus and spleen were calculated. The formation of serum hemolysin was determined by half hemolysis value method( HC50). After immunosuppressed mice model was induced by cyclophosphamide( CTX),the effects of TK on AST,ALT and BUN of mice serum were observed,and the thymus index and spleen index were calculated. Results: Acute toxicity testing showed that at the maximum drug concentration and the maximum drug delivery volume,TK mice grew well and no death occurred. Immune experiment showed that compared with control group,the phagocytic index αof mononuclear macrophage,HC50 and spleen index of TK group were significantly increased( P <0. 05). Compared with immunosuppressed mice model group induced by CTX,the content of AST,ALT and BUN of TK group were significantly decreased and spleen index significantly increased( P <0. 05). Conclusion: These results suggest that Semiaquilegia adoxoides extract( TK) can enhance immune function in mice,and to a certain extent can protect liver and kidney in mice from damage induced by CTX.
