摘要(Abstract):
目的:了解贵州省2008-2011年麻疹病毒(Measles Virus,MeV)分子流行病学特征,为控制和消除麻疹提供实验室依据。方法:将2008-2011年采集麻疹疑似病例的咽拭子或尿液共56份接种于Vero/SLAM(Signaling Lymphocyte Activation Molecule)细胞,培养获得8株阳性分离物,提取其病毒RNA,用逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)对麻疹病毒N基因羧基末端676个核苷酸片段进行扩增,对扩增产物进行序列测定和分析。结果:分离的8株MeV均为H1基因型中的H1a基因亚型,麻疹病毒分离株之间的核苷酸相似性为97.8%~100%,氨基酸相似性为96.0%~100%,与WHO H1基因型参考株Hunan.China93-7的核苷酸和氨基酸差异分别在2.0%~2.7%和2.0%~5.3%。结论:H1a基因亚型是贵州省近年来麻疹流行的优势基因型,未发现其他基因型。
Objective: To study molecular epidemiology characteristics of wild-type measles virus(MeV) isolated in Guizhou province during 2008-2011,and to provide lab evidence for measles control and elimination.Methods: Total 56 throat swabs and urine samples collected from suspected measles cases during 2008-2011 were inoculated into Vero/SLAM cells,and 8 strains of positive measles isolation were successfully obtained,and viral RNA was extracted.Fragments of 676 nucleotide acids of carboxyl terminal of measles viral nucleoprotein gene were amplified by using reverse transcription-polymerase chain reaction(RT-PCR) method,and then the PCR products were sequenced and analyzed.Results: All the 8 MeV strains were identified as H1a gene subtype.The similarity of their nucleotide was 97.8%~100%,and the similarity of their amino acid was 96.0%~100%.The differences of nucleotide and amino acid between 8 MeV strains and WHO H1 genotype reference strain: Hunan.China93-7 were 2.0%~2.7% and 2.0%~5.3% respectively.Conclusions: The dominant genotype of wild-type measles virus is H1a gene subtype in Guizhou province in recent years,other subtype is not found.
