MicroRNA-137在胰腺癌中的表达及临床意义

摘要(Abstract):

目的:检测MicroRNA-137(miR-137)在胰腺癌中的表达并探讨其临床意义。方法:采用实时荧光定量PCR(qRT-PCR)检测miR-137在17例临床胰腺癌组织及相配对的癌旁组织中的表达,同时,检测8株不同胰腺癌细胞系和2株正常胰腺上皮细胞中miR-137的表达,并分析miR-137的表达水平与胰腺癌临床病理参数之间的关系。结果:miR-137在胰腺癌组织中的表达水平明显低于癌旁组织,差异具有统计学意义(P<0.05);miR-137在胰腺癌细胞系中的表达水平明显低于正常胰腺上皮细胞(P<0.05);miR-137表达水平在临床Ⅰ~Ⅱ期胰腺癌组织明显高于Ⅲ~Ⅳ期胰腺癌组织(P<0.05),在无淋巴转移的胰腺癌组织明显高于有淋巴转移的胰腺癌组织(P<0.05)。结论:miR-137低表达与胰腺癌的发生发展相关,可能成为胰腺癌基因诊断和治疗的新靶点。
Objective: To detect microRNA-137( mi R-137) expression in pancreatic carcinoma,and to investigate its clinical significance. Methods: Real-time PCR( q RT-PCR) was used to detect mi R-137 expression in 17 cases of pancreatic carcinoma tissues and matched paracancerous tissues,8 pancreatic cancer cell lines and 2 normal pancreatic epithelial cell lines; the relationship between the expression level of mi R-137 in pancreatic cancer and pancreatic cancer clinical pathological parameters was analyzed. Results: Expression level of mi R-137 in pancreatic cancer tissue was significantly lower than that of adjacent tissues( P < 0. 05). Expression level of mi R-137 in pancreatic cancer cell lines was significantly lower than that in normal cell lines( P < 0. 05). The expression level of mi R-137 in the I ~ II stage pancreatic cancer tissue was significantly higher than that in Ⅲ ~ Ⅳ stage pancreatic carcinoma( P < 0. 05),that in pancreatic cancer without lymph node metastasis was significantly higher than that of pancreatic cancer with lymph node metastasis( P < 0. 05). Conclusion: Mi R-137 is related to the occurrence and development of pancreatic cancer. It may be a new target of gene diagnosis and therapy of pancreatic cancer.
