头花蓼内生真菌Diaporthe sp.代谢产物中的抗菌活性成分研究

摘要(Abstract):

目的:研究头花蓼内生真菌Diaporthe sp.代谢产物中的抗菌活性成分。方法:采用硅胶色谱、Sephadex LH-20及半制备高效液相色谱等分离方法对内生真菌Diaporthe sp.的代谢产物进行分离纯化,运用MS、1HNMR及13C-NMR等现代波谱分析技术对分离得到的单体化合物进行结构鉴定,采用96孔板二倍稀释法对单体化合物进行抗菌活性研究。结果:从内生菌代谢物中分离得到化合物11个,分别为胸腺嘧啶核苷(1)、2'-O-甲氧基尿嘧啶核苷(2)、尿嘧啶核苷(3)、对羟基苯甲醛(4)、alternariol(5)、2-甲基-3,5-羟基色酮(6)、尿嘧啶(7)、(-)-(3R)-mellein methylether(8)、3-(2R,4S-dihydroxypentyl)-6,8-dihydroxyisocoumarin(9)、2-(4-hydroxyphenyl)ethylacetate(10)和对羟基苯乙醇(11),化合物1~3和7~10为Diaporthe属首次报道;抗菌活性测试显示,化合物5、6、8及9对大肠杆菌的最低抑菌浓度(MIC)为16~32 mg/L,化合物5对金黄色葡萄球菌的MIC为32 mg/L。结论:化合物5、6、8及9为头花蓼内生真菌Diaporthe sp.代谢产物中的抗菌活性成分。
Objective: To study the antibacterial active components in the metabolites of endophytic fungi Diaporthe sp. of Polygonum capitatum. Methods: The metabolites of endophytic fungi Diaporthe sp. were isolated and purified by silica gel chromatography,Sephadex LH-20 and semi-prepared high performance liquid chromatography. MS,1 H-NMR,13 C-NMR and other modern spectral analysis techniques were used to identify the structure of the isolated monomer compounds. Antibacterial activity of single compound was studied by double dilution method with 96-well plate. Results: 11 compounds were isolated from endophytic metabolites: thymine nucleoside( 1),2'-O-methoxyuridine nucleoside( 2),uracil nucleoside( 3),p-hydroxybenzaldehyde( 4),alternariol( 5),2-methyl-3,5-hydroxytryptone( 6),uracil( 7),(-)-( 3 R)-melin methylether( 8),3-( 2 R,4 S-dihydroxypentyl)-6,8-dihydroxyisocoumarin( 9),2-( 4-hydroxyphenyl) ethylacetate( 10) and p-hydroxyphenylethanol( 11). The compounds 1-3 and 7-10 were first reported in Diaporthe genus. The antimicrobial activity test showed that the minimum inhibitory concentration( MIC) of compound 5,6,8 and 9 to Escherichia coli was 16-32 mg/L,and the MIC of compound 5 to Staphylococcus aureus was 32 mg/L. Conclusion: Compounds 5,6,8 and 9 are antimicrobial active components of endophytic fungi Diaporthe sp. metabolites of Polygonum capitatum.
