毛细管区带电泳法测定黄连、黄柏炮制前后小檗碱的含量
Berberine Contents in Coptis chinensis and Phellodenron chinense before and after Processing Tested with Capillary Zone Electrophoresis

摘要(Abstract):

目的:考察黄连、黄柏药材炮制前后小檗碱的含量变化。方法:采用熔融石英毛细管柱(50μm×72.5 cm,有效长度64.2 cm)作为分离通道,50 mmol/L Na2HPO4-NaH2PO4(pH=6.7)-甲醇(2∶1)作为缓冲溶液,运行电压30 kV,毛细管柱温20℃,检测波长275 nm。结果:小檗碱的线性范围为0.010 8~0.075 6 mg/L,r=0.999 3,小檗碱在黄连和黄柏中的回收率分别为101.56%、RSD 2.0%和95.6%、RSD 1.32%。结论:毛细管区带电泳法简便、准确,重复性好,黄连、黄柏炮制后小檗碱的含量均有降低。
Objective: To observe content changes of berberine in Coptis chinensis Franc and Phellodenron chinense Schneid before and after processing.Method: A fused quartz capillary(50 μm×72.5 cm,with effective length of 64.2 cm) was used as Abstraction tunnel.The running buffer was composed of 50mmol/L Na2HPO4-NaH2PO4(pH=6.7)-methanol(2∶1).The applied voltage was 30kv and the capillary temperature was 20℃.The detection wavelength was 275nm.Results: The linear curve for berberine was obtained in the range of 0.010 8 ~ 0.075 6 mg/L,r=0.999 3.The recovery of berberine in Coptis chinensis Franch was 101.56% with RSD of 2.0% and in Phellodenron chinense Schneid was 95.6% with RSD 1.32%.Conclusions: The method is simple,accurate and good in reproducibility.The berberine contents in Coptis chinensis Franc and Phellodenron chinense Schneid decrease after processing.
