CHIRALPAK AD-RH手性柱直接拆分普罗帕酮对映体
Direct Chiral Separation of Propafenone Enantiomers by RP-HPLC Using Chiralpak AD-RH

摘要(Abstract):

目的:建立普罗帕酮对映体的反相高效液相拆分方法,为研究普罗帕酮手性异构体药代动力学提供依据。方法:在Chiralpak AD-RH柱上,考察流动相pH值、流速、乙腈比例、缓冲盐浓度、柱温对普罗帕酮手性异构体拆分的影响。结果:以20mmol/L磷酸盐缓冲液(pH8.5)-乙腈为流动相,流速0.5mmol/L,柱温25℃,在Chiralpak AD-RH手性柱上成功拆分普罗帕酮对映体。结论:普罗帕酮对映体在Chiralpak AD-RH手性柱上获得基线拆分。
Objective:To develop a RP-HPLC method for the enantiomeric separation of propafenone on a new chiral column Chiralpak AD-RH [amylose tris (3,5-dimethylphenylcarbamate) coated on a 5 μm silica-gel substrate] for pharmaceutical study of propafenone. Mehtods: Enantiomeric separation of propafenone was carried out with Chiralpak AD-RH,and the influences of mobile phase pH value,flow rate,ratio of acetonitrile,concentration of buffer,and column temperature on the chiral separation were observed. Results: Propafenone enantiomers were successfully separated on Chiralpak AD-RH with a mobile phase consisting of 20 mmol/L phosphate buffer (pH8.5) and 1∶1 (V/V) acetonitrile at a flow rate of 0.5 mmol/L. Conclusion: A baseline separation of propafenone enantiomers was achieved on a Chiralpak AD-RH column.
