氟维司群对雌激素诱导的MCF-7乳腺癌细胞侵袭及基质金属蛋白酶表达的影响The Effects of Fulvestrant on Invasion Ability and Matrix Metalloproteinase Expression Induced by Estrogen in Breast Cancer Cell MCF-7

摘要(Abstract):
目的:观察氟维司群(ICI)对雌激素(E2)诱导的人乳腺癌细胞(MCF-7)的迁移能力、侵袭能力及基质金属蛋白酶MMP2、MMP9蛋白表达的影响。方法:将对数生长期的MCF-7细胞分为对照组、ICI组、E2组和(ICI+E2)组,采用伤口愈合实验检测MCF-7细胞迁移率,基质胶包被的transwell小室实验检测细胞的侵袭能力,western-blot法检测MMP2、MMP9的蛋白表达,实时荧光定量PCR检测MMP9 mRNA的水平。结果:(1)与对照组比较,E2组、ICI组及(ICI+E2)组使MCF-7细胞的迁移和侵袭能力增强(P<0.05);与E2组比较,(ICI+E2)组不能使MCF-7细胞的侵袭能力增强(P>0.05);(2)与对照组比较,E2组、ICI组及(ICI+E2)组对MCF-7细胞中MMP2蛋白水平的影响不明显(P>0.05),但可提高MCF-7细胞中MMP9蛋白的水平(P<0.05);(3)与对照组比较,E2组、ICI组及(ICI+E2)组MCF-7细胞MMP9 mRNA增加(P<0.05)。结论:ICI不能抑制E2诱导的乳腺癌细胞的迁移和侵袭,单独作用时可增加细胞的迁移、侵袭能力,这可能与其增加MMP9表达有关。
Objective: To observe the effects of fulvestrant( ICI) on estrogen( E2)-induced migration,invasion and the expression of MMP2,MMP9 in breast cancer cell MCF-7. Methods: MCF-7cells in logarithmic growth period were divided into control group,E2 group,ICI group and E2 + ICI group. Cell migration was determined by wound healing assay. Cell invasion was determined with matrigel coated transwell assay. MMP2 and MMP9 protein expression was determined by western-blot.The mRNA levels of MMP9 was examined by real time fluorescence quantitative PCR( real-time PCR). Results: Compared with control group,migration and invasion abilities of MCF-7 cells were increased in the other three groups( P < 0. 05). Compared with group E2,migration and invasion abilities of MCF-7 cells were increased in group E2 + ICI( P < 0. 05). Compared with control group,the expression of MMP2 protein showed no obvious change in the other three groups,but the expression of MMP7 protein was increased in the three groups,and that in in group E2 + ICI was higher than that in group E2( P < 0. 05). Compared with control group,the expression of MMP7 mRNA was increased in the three groups( P < 0. 05). Conclusions: ICI can not inhibit the migration and invasion of breast cancer cells induced by E2,and ICI alone can increase cell migration and invasion,which may be related to increase of MMP9 protein expression.
