胶原诱导性关节炎小鼠脾脏树突状细胞的检测及意义

摘要(Abstract):

目的:观察胶原诱导性关节炎(CIA)小鼠病程中脾脏树突状细胞(DC)亚群的比例及变化规律。方法:将6~8周龄近交系雄性DBA/l小鼠12只分为对照组(n=6)和模型组(n=6),模型组小鼠参照经典的CIA模型方法制备CIA模型,正常组小鼠同剂量尾根部注射0. 9%氯化钠溶液;观察两组小鼠关节情况,用关节炎指数(AI)评分衡量关节病变情况,采用HE染色观察关节的组织形态学改变,通过流式细胞术检测脾经典DC(cDC)及CD8~+cDC、CD4~+CD8~-cDC、CD4~-CD8~-DC各亚群的变化。结果:与对照组比较,模型组小鼠出现明显的关节红肿、行动迟缓等表现,HE染色显示模型组小鼠踝关节结构被破坏,关节软骨可见明显坏死脱落、纤维化,并有大量炎细胞浸润,且AI评分高于对照组(P <0. 05),脾脏cDC及CD4~+CD8~-cDC比例降低(P <0. 05),CD4~-CD8~-cDC比例升高(P <0. 05)。结论:CIA小鼠脾脏DC细胞亚群的变化可能与RA病情的进展有关。
Objective: To observe the proportion and variation of splenic dendritic cell( DC) subsets in the course of collagen induced arthritis( CIA) mice. Methods: The male DBA/l mice of 6-8 weeks were divided into normal group( n = 6) and model group( n = 6). The CIA model was prepared according to the classical CIA model method. The normal mice were injected with 0. 9% sodium chloride solution at the same dose of tail root. The joint condition of two groups was observed,the joint lesion was measured by arthritis index( AI) score,the histomorphologic changes of the joint were observed by HE staining,and the changes of splenic classical DC( cDC) and CD8~+cDC,CD4~+CD8~-cDC,CD4~-CD8~-DC subsets were detected by flow cytometry. Results: Compared with the control group,the mice in the model group showed obvious joint redness and retardation. HE staining showed that the ankle joint structure of the model group was destroyed,and the articular cartilage showed obvious necrosis,fibrosis,and a large number of inflammatory cell infiltration. AI score was higher than control group( P < 0. 05). The ratio of spleen cDC and CD4~+CD8~-cDC decreased( P < 0. 05).The ratio of CD4~-CD8~-cDC increased( P < 0. 05). Conclusion: The changes of spleen DC cell subsets in CIA mice may be related to the progression of RA disease.
