星形胶质细胞α7神经型尼古丁受体激动剂Nicotine和PNU对钙调蛋白水平的影响The Influence of Nicotine and PNU on the Expression of Calmodulin in Astrocytes of SD Rats

摘要(Abstract):
目的:探讨星形胶质细胞α7神经型尼古丁受体(α7nAChRs)激动剂Nicotine和PNU对钙调蛋白(CaM)表达水平的影响。方法:第3~4代乳鼠大脑星形胶质细胞分为对照组和实验组,对照组不加α7nAChRs激动剂,实验组分别加入α7nAChRs激动剂Nicotine或PNU处理细胞,Nicotine或PNU的浓度为1、5及10μmol/L,处理时间为6、12、18及24 h;采用蛋白印迹(Western blotting)方法检测Nicotine或PNU不同浓度及不同时间点细胞裂解液中的CaM蛋白水平。结果:5μmol/L Nicotine处理6、12、18及24 h和10μmol/L Nicotine处理12、18及24 h的星形胶质细胞裂解液中的CaM蛋白表达水平高于对照组(P<0.05),其余Nicotine浓度和处理时间点星形胶质细胞裂解液中CaM蛋白表达水平虽略高于对照组,但差异无统计学意义(P>0.05);浓度为1、10μmol/L的PNU处理24 h,CaM蛋白表达与对照组比较差异无统计学意义(P>0.05),其他浓度及时间点的CaM蛋白表达水平均高于对照组(P<0.05)。结论 :星形胶质细胞α7 nAChRs的激活可以使CaM的蛋白表达水平增加。
Objective: To investigate the influence of activators of alpha7 cholinergic receptor(α7 nAChRs),nicotine and PNU,on the expression of calmodulin( CaM) in astrocytes. Methods:The 3-4 generations of cultivated astrocytes were divided into normal control group( group C) and experiment group( group E). Cells in group C did not receive treatment of nicotine,or PNU. In group E,cells were treated with 1,5,and 10 μmol/L respectively of nicotine or PNU for 6 h,12 h,18 h and 24 h. The expression level of CaM was detected by using western blotting method. Results: In group E,when cells were treated by nicotine at 5 μmol/L for 6,12,18 and 24 h,and at 10 μmol/L for 12,18,and 24 h,the expression level of CaM was higher than that in group C. While for nicotine at other doses and other treating time points,the CaM expression was some what higher than that of group C,but the difference was not significant statistically( P > 0. 05). When cells were treated by PNU at doses of 1 and 10 μmol/L for 24 h,the CaM expression was not different from that in group C( P > 0. 05),while at other doses of PNU and other treating time points,the CaM expression was higher than that in group C( P < 0. 05). Conclusion: The activation of astrocytic α7 nAChRs leads to the increase of protein expression levels of CaM.
