T细胞表面分子CD28及其信号通路的研究

摘要(Abstract):

用免疫磁性阴极选择法从正常人外周血单个核细胞中择取 T 细胞,经 CD28单克隆抗体和(或)乙酰肉豆蔻佛波酯刺激,Western 印渍免疫沉淀反应,白细胞介素-2和白细胞介素-2受体等检测,结果表明 CD28单抗和乙酰肉豆蔻佛波酯复合刺激具有诱导 c-Rel 核内移位和磷酸化、增强白细胞介素-2受体表达和白细胞介素-2分泌的作用,证明 CD28信号途径有诱导环孢素 A 抵抗-非钙依赖和环孢素A敏感-钙依赖的传导通路,对 T 细胞活化有重要意义。
CD28 is one of surface accessory molecules on T cells.CD28 receptor stimulation is requred for T cell responses to antigens and for B cell responses to T-dependent antigens.CD28 ligation induced tyrosine phosphorylation of specific substrates and is a noval signaling transduction pathway for reg- ulating T cell activation.T cells enriched by immunomagnetic selection from PBMC were stimulated or costimulated with anti-CD28 mAb/PMA.Using the methods of western bloting,immunoprecipi- tation,IL-2 and IL-2R assay showed that the costimulation of CD28 with PMA induced transloeation of NF-kB family proteins and their tyrosine phosphorylation and increased the expression of IL-2R and IL-2 secretion.The results demonstrate the important role of CD28 receptor for inducing resistant-cal- cium independent and CsA seusitive-calcium dependent transduction pathway during T cell responses to antigen.
