用登革Ⅲ型病毒免疫小鼠后特异性IgG抗体的动态变化

摘要(Abstract):

目的 :以登革Ⅲ型病毒 (DV3 )免疫成年BALB/C小鼠 ,观察血液中特异性IgG抗体的动态变化。 方法 :以不同剂量DV3 经皮下注射和肌肉注射接种成年BALB/C小鼠 ,用间接免疫荧光法检测不同时间机体末梢血中特异性IgG抗体及其水平。 结果 :不同剂量DV3 免疫BALB/C小鼠均可诱导体液免疫应答 ,特异性IgG抗体于免疫后第 8~ 12天开始产生 ,于第 15~ 18天达高峰 (1∶80 ) ,继而下降。特异性IgG类抗体可维持 47d以上 ,其中以 10 0 0TCID50 DV3 免疫BALB/C小鼠最早产生特异性抗体且水平最高。结论 :经皮下和肌肉注射DV3 ,可诱导BALB/C小鼠产生特异性IgG类抗体。
Objective:The specific IgG antibody changes were observed in the serum of BALB/C mice immunized with dengue 3 virus(DV3). Methods: Male BALB/C mice were immunized with different volumes of DV 3 by subintradermal and intramuscular injections. The level of specific anti DV 3 IgG antibody in serum was examined by using an indirect immunofluorescence assay. Results: The BALB/C mice immunized with DV 3 of different volumes produced humoral immunity. The specific IgG antibody was generated 8-12 days after immunization and the top level appeared in 15-18 days(1∶80). 21 days after infection, the antibody level gradually decreased, and it could still be detected 47 days after being immunized. Conclusion: Specific IgG antibody could be induced in BALB/C mice immunized with Dengue 3 virus by subintradermal and intramuscular injections.
