HPLC测定不同采收期水黄花中东莨菪内酯、金丝桃苷及槲皮苷含量Content Determination of 3 Compounds in Euphorbia chrysocoma Harvested during Different Period by HPLC

摘要(Abstract):
目的:建立水黄花中东莨菪内酯、金丝桃苷、槲皮苷的含量测定方法,为评价水黄花的最佳采收期提供依据。方法:采用高效液相色谱法对不同时间点采自贵阳市郊区的22批水黄花中的东莨菪碱、金丝桃苷及槲皮苷进行检测,色谱柱Phenomenon Luna C18柱(4.6 mm×250 mm,5μm),柱温为40℃,流速为1.0 m L/min,检测波长为360 nm,流动相为乙腈-0.2%磷酸水,梯度洗脱;测定22批不同采收期的水黄花中3种成分的含量。结果:东莨菪内酯在2.54~254.0 mg/L(r=0.999 3)、金丝桃苷在0.67~67.0 mg/L(r=0.999 5)、槲皮苷在1.10~110 mg/L(r=0.999 5)的进样浓度范围内线性关系良好,加样回收率为98.72%~100.0%,RSD小于3%,22批药材中3种被测成分含量差距较大,9~11月采集的水黄花中3种成分含量较高。结论:建立了水黄花药材中3种主要成分东莨菪碱、金丝桃苷及槲皮苷含量测定方法,9~11月采集的水黄花中的3种主要成分含量较高。
Objective: To set up a method for simultaneous content determining of scopoletin,hyperoside,quercetin in Euphorbia chrysocoma,and provide a reference basis to evaluate the best harvest period of the herb. Methods: Contents of scopoletin,hyperoside,quercetin in 22 batches of Euphorbia chrysocoma from Guiyang collected at different time points were tested by using HPLC. The column used was Phenomenon Luna C18(4.6 mm × 250 mm,5 μm),column temperature 40 ℃,flow speed1. 0 m L/min,determining wave length 360 nm,and the mobile phase acetonitrile-0.02 % phosphoric acid. Gradient elution was conducted. Result: Good linear relationships of scopoletin,hyperoside and quercetin was showed within the ranges of 2. 54 ~ 254. 0 mg/L(r=0.9993),0. 67 ~ 67. 0 mg/L(r>0.9995) and 1. 10 ~ 110 mg/L(r>0.9995) respectively. The recovery rate of 3 measured compounds were in the range of 98. 72 % and 100. 0 %,and each RSD was under 3 %(n=6). The contents of 3 measured compounds showed large differences in 22 batches of determined herbs.Conclusion: Contents of the 3 kinds of compounds relate to the collection seasons. Herbs gathered during September through November have higher levels of the 3 compounds.
