不同途径感染HSV-Ⅱ后小鼠脑及脊髓病理损伤及易受累组织的病毒载量
The Difference Caused by Different Routes of Infection of Herpes Simplex Virus Type 2 in Mice and the Pathological Damage and Distribution of Tissue Viral Load

摘要(Abstract):

目的:分析不同途径感染Ⅱ型单纯疱疹病毒(HSV-Ⅱ)对小鼠的病理损伤及易受累组织的病毒载量。方法:60只清洁级成年BALB/c小鼠随机分为5组,实验A、B、C、D组分别在小鼠眼角膜、鼻黏膜、生殖道及背部皮下接种HSV-Ⅱ,对照组经皮下注射PBS;实验第1~15天,每组各取4只小鼠,观察小鼠临床症状、体质量及生存率;实验第3天及第6天取实验组小鼠2只,观察小鼠脑及脊髓病理损伤,脑、脊髓、三叉神经节及生殖道组织的病毒载量。结果:与对照组相比,实验组临床症状分级增高,由高到低为C组、B组、A组、D组,实验A、B、C组体质量下降幅度较大,实验D组体质量下降不大;实验A、B、C组小鼠,有较高致死率,分别为50%、25%及25%;实验A组及B组脊髓组织中可见部分神经细胞被噬,而实验C组与D组神经细胞吞噬现象不明显;实验A组与实验B组小鼠的脑组织、脊髓组织、三叉神经节组织病毒载量最高,而实验C组生殖道组织病毒载量最高,实验D组小鼠各组织病毒载量较其他实验组低。结论:小鼠感染HSV-Ⅱ后脑及脊髓病理损伤情况及病毒载量与感染途径有关,经眼角膜、鼻黏膜、生殖道感染HSV-Ⅱ的小鼠病理损伤及病毒载量较经皮感染严重。
Objective: To analyze the pathological damage and viral load distribution of mice infected with herpes simplex virus type 2( HSV-Ⅱ) caused by different routes. Methods: 60 adult BALB/c mice of clean grade were randomly divided into 5 groups. The control group received subcutaneous injection of phosphate buffered saline( PBS) as pseudo-inoculation. The other 4 groups were vaccinated with HSV-Ⅱ in the cornea,nasal mucosa,genital tract and dorsal subcutaneous tissues respectively,so as to to compare the clinical symptoms,pathological damage,and viral load among the mouse model groups. Results: The HSV-Ⅱ vaccinated 4 groups of mice exhibited abnormalities such as arched back,inverted hair,paralysis of the hind limbs,weight loss,suppuration and other symptoms were all compared with those of the control group; In the period of observation,the mortality rate of the mice with nasal mucosa infection was 50%,while 25 % of the mice with cornea and genital tract infection died. No infection was found in the mice with back infection. Pathological examination showed that there were hemorrhage and inflammatory cells in the brain and spinal cord tissues of the mice. The cornea and nasal mucosa groups were severely damaged. High levels of viral loads were detected in the brain,spinal cord and trigeminal ganglion tissues. Conclusion: This study may provide a scientific basis for the research and development of clinical vaccine strains and attenuated strains in the awareness of the pathological damages and the distribution of viral loads of the mice with HSV-Ⅱ infected through different pathways.
