摘要(Abstract):
目的:用明胶海绵吸收法回收DNA片段,并验证该方法能否有效、快速的从电泳后琼脂糖凝胶中回收DNA片段。方法:将明胶海绵剪成楔形,在紫外灯照射下插入含有目的DNA片段的琼脂糖凝胶中,待海绵完全膨胀后取出,离心收集DNA溶液;并用该法回收的DNA片段进行PCR扩增,检验其回收的DNA质量。结果:明胶海绵吸收法回收DNA样品的得率介于63%~98%,所回收的DNA进行PCR扩增能够获得相应的目的条带。结论:明胶海绵吸收法是一种快速、有效的DNA回收方法。
Objective:To develop a novel method for DNA recovery,and verify its feasibility in extraction and purification of DNA segments from agarose gel after electrophoresis. Methods:A piece of gelatin sponge was cut to a wedge-form stick and inserted into agarose gel at place of goal DNA fragment under long-wavelength UV. DNA solution was collected by centrifuging after the sponge was fully swelled. The amount and purity of DNA sample were determined by PCR. Results:DNA yield rates were in the range of 63~98%,and the target DNA fragments amplified by PCR were intact. Conclusion:The sponge absorption method is feasible,effective and fast for DNA recovery.
