S100A8/A9蛋白对人舌鳞癌SCC-25及CAL-27细胞侵袭的影响

摘要(Abstract):

目的:探讨外源性S100A8/A9蛋白对人舌鳞癌细胞SCC-25及CAL-27侵袭能力的影响。方法:人舌鳞癌SCC-25及CAL-27细胞分别分为实验组及对照组,实验组用5 mg/L S100A8/A9蛋白干预24 h,对照组添加等量培养基;采用Transwell小室侵袭实验检测SCC-25及CAL-27细胞侵袭情况,实时定量PCR(RT-qPCR)检测SCC-25及CAL-27细胞中基质金属蛋白酶-2(MMP-2)及MMP-9基因的表达情况。结果:与对照组比较,实验组SCC-25及CAL-27细胞在Transwell小室中降解人工基底膜并穿过聚碳酸酯膜的能力增强(P<0.05),且实验组SCC-25及CAL-27细胞MMP-2及MMP-9的基因相对表达量增加(P<0.05)。结论:5 mg/L外源性S100A8/A9蛋白能增强舌癌SCC-25及CAL-27细胞的侵袭能力,其机制可能与S100A8/A9蛋白影响两种细胞中MMP-2及MMP-9表达有关。
Objective: To investigate the effect of exogenous S100 A8/A9 protein on the invasive ability of human tongue squamous cell carcinoma(TSCC) SCC-25 and CAL-27 and to provide a theoretical basis for the study of the mechanism of invasion and metastasis of S100 A8/A9 protein in tongue squamous cell carcinoma. Methods: Human tongue squamous cell carcinoma scc-25 and cal-27 cells were divided into the experimental group and the control group respectively. The experimental group was treated with 5 mg/L S100 A8/A9 protein for 24 h, and the control group was treated with the same amount of medium.Transwell cell invasion assay was used to detect the invasion of S100 A8/A9 protein on SCC-25 and CAL-27 cells after 24 hours. RT-q PCR was used to detect the expression of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) genes in SCC-25 and CAL-27 cells. Results: Compared with the control group, adding 5 mg/L exogenous S100 A8/A9 protein could enhance the ability of SCC-25 and CAL-27 cells of the experimental group to degrade artificial basement membrane and pass through polycarbonate membrane in Transwell chamber(P<0.05),and the relative expression of MMP-2 and MMP-9 genes was increased(P<0.05). Conclusion: 5 mg/L exogenous S100 A8/A9 protein can enhance the invasion ability of tongue cancer scc-25 and cal-27 cells, and the mechanism may be related to the influence of S100 A8/A9 protein on the expression of mmp-2 and mmp-9,which may provide a potential biomarker for targeted therapy of tongue squamous cell carcinoma.
