摘要(Abstract):
目的:探究钙结合蛋白S100A4对鸡卵清白蛋白(OVA)诱导的免疫应答作用。方法:10只C57BL/6雌性野生型(WT)小鼠随机均分OVA组(右踝关节注射OVA 20μg)与OVA联合S100A4蛋白组(右踝关节注射OVA 20μg和S100A4蛋白5μg混合液,即联合组),注射2周后收集2组小鼠引流淋巴结和血清,采用流式细胞术检测引流淋巴结T细胞与树突状细胞(DC)及其活化百分比,采用酶联免疫吸附测定法(ELISA)检测血清中OVA特异性免疫球蛋白G(Ig G)和免疫球蛋白E(Ig E)的水平。结果:流式细胞术检测结果显示,联合组小鼠引流淋巴结中T细胞(CD3~+)、DC(CD11c~+)、活化T细胞(CD3~+CD69~+)及活化DC(CD11c~+MHC-Ⅱ~+)百分比均高于OVA组(P <0. 05); ELISA检测结果显示,联合组小鼠血清中OVA特异性Ig G明显高于OVA组(P <0. 01)。结论:S100A4蛋白能够促进OVA诱导的免疫应答,起着免疫佐剂的作用。
Objective: To investigate the role of calcium-binding protein S100 A4 in the immune response induced by Ovalbumin( OVA). Methods: 10 mice of C57 BL/6 female wild-type( WT)were randomly divided into OVA group( with 20 μg of OVA injected into the right hock) and Joint group( with 20 μg of OVA and 5 μg of S100 A4 protein injected into the right hock). Two weeks later,the draining lymph nodes and serum of the two groups were collected,and the frequencies of T cells and dendritic cells( DC) in draining lymph nodes and their activation frequencies were detected by flow cytometry. The levels of immunoglobulin G( IgG) and immunoglobulin E( IgE) in serum was measured by enzyme-linked immunosorbent assay( ELISA). Results: The Flow Cytometry showed that the frequencies of T cells( CD3~+),dendritic cells( CD11 c~+),activating T cells( CD3~+CD69~+),and activating dendritic cells( CD11 c~+MHC-Ⅱ~+) in Joint group were significantly higher than those in OVA group( P < 0. 05). ELISA showed that the IgG level in Joint group was significantly higher than in OVA group( P < 0. 01). Conclusion: S100 A4 protein can act as an immunologic adjuvant and promote the immune responses induced by OVA.
