摘要(Abstract):
目的:探讨TGF-β1对体外培养人视网膜色素上皮(human retinal pigment epithelium,hRPE)细胞增殖率及I型胶原表达的影响。方法:实验组用0.1、1、10μg/L的TGF-β1对体外培养hRPE细胞进行干预,对照组只加培养液培养hRPE细胞,用RT-PCR与酶联免疫吸附法检测Ⅰ型胶原mRNA和蛋白表达,用MTT法检测细胞的增殖率。结果:实验组hRPE细胞Ⅰ型胶原的mRNA和蛋白表达量及细胞增殖率较对照组明显增高(P<0.05),1μg/L组进一步升高(P<0.05),10μg/L组达高峰(P<0.05),呈现明显的剂量依赖性,TGF-β1浓度与Ⅰ型胶原蛋白的表达及增殖率的相关系数r分别为0.863及0.901。结论:TGF-β1能够促进hRPE细胞Ⅰ型胶原的表达和细胞的增殖,这种现象与TGF-β1的浓度成正相关。
Objective: To explore the effects of transforming growth factor beta 1(TGF-β1) on the proliferation of cultured human retinal pigment epithelium(hRPE) cells and expression of type I collagen.Methods: TGF-β1 at concentrations of 0.1,1,and 10 μg/L was added into cultures of hRPE cells in experimental groups respectively(groups 0.1,1 and 10),and in the control group of hRPE cell culture,no TGF-β1 was added.Expressions of mRNA and protein of type Ⅰcollagen were detected by using RT-PCR and enzyme linked immunosorbent assay,and proliferation of hRPE cells was detected with MTT.Results: The proliferation and expression of mRNA and protein of typeⅠ collagen in group 0.1 were significantly higher than those in control group(P<0.05),and they were getting much higher in group 1(P<0.05).In group 10,they reached the peak(P<0.05),and a dose-dependent increase manner displayed.The correlation coefficients of TGF-β 1concentrations with type Ⅰcollagen protein expression and proliferation rate were r=0.863 and r=0.901 respectively.Conclusion: TGF-β1 at different concentrations can promote the expression of type Ⅰ collagen protein and hRPE proliferation.There is a positive correlation between TGF-β1 concentration and the expression of type Ⅰ collagen,and between TGF-β1 concentration and hRPE proliferation.
