摘要(Abstract):
目的:研究环维黄杨星D(CVB-D)对Na2S2O4诱导EVC304细胞损伤的保护作用。方法:采用体外培养人脐静脉内皮细胞EVC304,Na2S2O4作为耗氧剂建立细胞缺氧损伤模型。用MTT法测定细胞存活率与细胞培养液乳酸脱氢酶(LDH)活力反映细胞损伤的程度,培养液一氧化氮合酶(NOS)活力与内皮素(ET)含量分析EVC304细胞的功能。结果:CVB-D能显著抑制Na2S2O4诱导EVC304细胞OD570的降低和细胞培养液中LDH活力的增加,提高培养液中NOS活性,降低ET的含量(与模型组比较,P<0.05或P<0.01)。结论:CVB-D对Na2S2O4诱导EVC304细胞损伤具有保护作用,其作用机制可能与改善NOS和ET失调有关。
Objective:To investigate the protective effects of cyclovirobuxine D (CVB-D)on human umbilical vein endothelial cell (EVC304) from the injury induced by Na2S2O4 in vitro. Methods:Cell injury model of cultured EVC304 was induced by Na2S2O4 as oxygen comsuming agent. Cell injury degree was determined with methyl thiazolyl tetrazolium (MTT) stainning method.and lactate dehydrogenase (LDH) activity in culturing medium. Cellular function of EVC304 was indicated by nitric oxide syntyhase (NOS) activity and endothelin (ET) content in the medium. Results:Cell survival ratio and NOS activity significantly decreased after treated with Na2S2O4,and the LDH activity and ET concents significantly increased (P<0.05,P<0.01). CVB-D remarkablely attenuated EVC304 injury induced by Na2S2O4. Conclusions:CVB-D can protect ECV304 from injury induced by Na2S2O4. The protective mechanism may relate to ameliorating disturbance of NOS and ET of ECV304.
