局部注射丹参酮Ⅱ-A对兔正畸牙移动中破骨细胞的影响
The Impact of Local Injection of Tanshinone Ⅱ-A on Osteoclast of Orthodontic Tooth of Rabbit

摘要(Abstract):

目的:观察局部注射丹参酮Ⅱ-A对家兔正畸牙移动过程中牙周膜压力侧破骨细胞的影响。方法:选取20只雄性健康大耳白兔复制双侧下颌第一磨牙近中移动模型,左侧下颌第一磨牙近中牙龈黏膜下注射丹参酮Ⅱ-A磺酸钠注射液0.36 mg/d,作为实验组,对侧同名牙位处注射等量生理盐水作为对照组,于实验第1、3、7、14及21天时处死兔,取下颌印模灌石膏模型并测量双侧下颌第一磨牙近中移动距离;取兔双侧下颌第一磨牙和周围牙槽骨组织,行抗酒石酸酸性磷酸酶(TRAP)染色,光镜下计数牙根中1/3压力侧牙周膜及牙槽骨表面破骨细胞数量。结果:除实验第1天外,实验组其余各组兔牙移动距离大于对照组,差异有统计学意义(P<0.05);两组兔牙周膜压力侧破骨细胞数量均有增加,实验组破骨细胞数量明显高于对照组,差异有统计学意义(P<0.05)。结论:牙移动过程中,适量丹参酮II-A可以有效的增加破骨细胞的数量,增强细胞活性,提高骨吸收强度,对加速正畸牙移动有一定的作用。
Objective: To observe the effect of local inject tanshinone Ⅱ-A on the osteoclast in the pressure side of periodontal membrane during orthodontic tooth movement. Methods: Twenty healthy and male rabbits were randomly divided into 5 groups( 4 rabbits each group) to copy the bilateral mandibular first molar movement model. 0. 36 mg / d tanshinone II A sulfonate was injected into the gingival mucosa of mesial surface of the left first molar once a day as experimental group while the same amount of normal saline was injected into the opposite molar of the same name as control group. The rabbits were killed at 1th,3th,7th,14 th,21thday after injection. The distance of bilateral mandibular first molar mesial movement was measured by plaster cast. The periodontal slice of the mandibular first molar was taken and conducted by HE staining and tartrate resistant acid phosphatase staining. The osteoclasts in the pressure side of periodontal membrane were counted under the light microscope. Results: Except for the frist day,the tooth movement distance of experimental group were significantly higher than that of control group( P < 0. 05). Compared with control group,the number of osteoclasts significantly increased in experimental group( P < 0. 05). Conclusion: In the process of orthodontic tooth movement,tanshinone Ⅱ-A can increase the number of osteoclasts,enhance cell activity and bone resorption intensity and improve the tooth movement.
