柴胡疏肝散对抑郁大鼠海马微管相关蛋白-2磷酸化的影响Influence of Bupleurum Liver-Coursing Powder on Phosphorylatied Microtubule-Associated Protein-2 in Hippocampus of Depressive Rats

摘要(Abstract):
目的:探讨柴胡疏肝散对抑郁大鼠海马组织微管相关蛋白-2(MAP-2)磷酸化的影响。方法:成年雄性Wistar大鼠45只,随机均分为对照组、模型组和治疗组;模型组和治疗组采用慢性不可预性温和应激建立抑郁症大鼠模型,治疗组同时经灌胃给予柴胡疏肝散治疗,持续22 d;采用糖水实验、高架十字迷宫实验和Morris水迷宫实验观察各组大鼠行为学变化,Western blot方法检测各组大鼠海马组织MAP-2及磷酸化MAP-2(p MAP-2)表达,Real-time PCR检测p MAP-2 mRNA表达。结果:与对照组比较,模型组大鼠糖水偏好百分比明显降低,进入开臂次数和停留时间百分比增高,水迷宫逃避潜伏期延长,海马p MAP-2蛋白和mRNA表达水平明显降低,差异均有统计学意义(P<0.05);与模型组比较,治疗组大鼠糖水偏好百分比明显增高,进入开臂次数和停留时间百分比降低,水迷宫逃避潜伏期减少,海马p MAP-2蛋白和mRNA表达水平明显增高,差异均有统计学意义(P<0.05)。结论:柴胡疏肝散具有显著的抗抑郁作用,该作用可能与影响海马MAP-2磷酸化有关。
Objective: To observe the influence of bupleurum liver-coursing powder on phosphorylation of microtubule-associated protein 2( p MAP-2) in hippocampus of depressive rats. Methods: Forty-five male Wistar rats were randomly divided into control group,model group and treatment group.The depression animal model was produced by giving the rats chronic unpredicted mild stress and treatment group received intragastric administration of bupleurum liver-coursing powder for 22 days. The sucrose preference test,elevated plus maze test and Morris water maze test were adopted to observe behavioral changes of rats in each group. The expression of p MAP-2 protein and mRNA were detected by western blot and real-time PCR respectively. Results: Compared with control group,sucrose preference percentage of model group was significantly lower,the number of entry into open arms and percentage of detention time extended significantly,the escape latency of Morris water maze test increased significantly,and the expression of p MAP-2 protein and mRNA were decreased significantly( P <0. 05). Compared with model group,sucrose preference percentage of treatment was significantly higher,the number of entry into open arms and percentage of detention time decreased significantly,the escape latency of Morris water maze test decreased significantly,and the expression of p MAP-2 protein and mRNA were increased significantly( P < 0. 05). Conclusion: Bupleurum liver-coursing powder has a significant anti-depressant effect,and its mechanism may be relavent to its effect on the phosphorylation of MAP-2 in hippocampus.
