体外共刺激阻断模型中NK细胞功能抑制机制初探A Preliminary Research on the Inhibition Mechanism to NK Cell in vitro with Co-stimulating-blocking Models

摘要(Abstract):
目的: 研究耐受状态下NK细胞功能抑制的机制。方法:利用anti-CD80抗体体外阻断CD28 /B7共刺激通路T细胞耐受模型,以51Cr标记YAC-1细胞为NK靶细胞,作51Cr标准4h释放实验,观察耐受T细胞与NK细胞共培养后对NK细胞的抑制效应。结果:51Cr释放率,耐受诱导组21%,抗原激活组24 4%,细胞对照组34%,无T细胞对照组32%;前3组上清液与NK细胞作用后靶细胞51Cr释放率分别为34%、31 6%、33 1%, 1、2两组51Cr释放率显著低于其余各组(P<0 01),而两组之间无显著性差异;无T细胞对照组与各组上清液中51Cr释放率无显著性差异。结论:在实验条件下, (1)抗原激活的T细胞及共刺激阻断后耐受的T细胞可显著抑制Balb/c小鼠NK细胞的细胞毒活性,且二者相比无显著性差异; (2)耐受T细胞与抗原反应性T细胞或未经抗原刺激的T细胞培养上清液对NK细胞的影响无显著性差异。根据体外实验结果,可初步认为T细胞可能主要以细胞间接触的方式来调节NK细胞的功能。
Objective: To investigate the inhibition mechanism of T cells under tolerant condition to NK cell functions. Methods: CD28/B7 passage of co-stimulating-blocking model of T cells under to-lerant condition was blocked by anti-CD80 antibody in vitro. YAC-1 cells were labeled with 51 Cr as the target of NK cells. 51 Cr standard 4h release experiment was conducted. The inhibiting effect on NK cells were observed after the co-incubation of tolerant T cells and NK cells. Results: The release rates of 51 Cr were 21%, 24.4%, 34% and 31.6% in tolerance-induced group (TI), antigen-activated group (AA), cell control group (CC), and cell control group without T cells (NT) respectively. In TI, AA and CC groups, the 51 Cr release rates of target cells were 34%, 31.6%, and 33.1% respectively, after the incubation of supernates with NK cells. the 51 Cr release rates of TI and AA groups were lower than those of the other groups (P<0.01). No significant difference was found between the 51 Cr release rates of TI and AA groups or among group NT and supernates of other groups. Conclusions: Under the condition of this experiment, 1) Allo-antigen activated T cell and co-stimulating-blocking T cells under tolerant condition can inhibit cytotoxic activity of NK cells of Balb/c mice, and there is no obvious difference between the 2 kinds of T cells; 2) There is no significant difference between the effects of incubation supernates of tolerant T cell, antigen-reacting T cell, and non-antigen-simulated T cell to NK cell. According to the results of the experiment in vitro, it seems that T cell might regulate functions of NK cell mainly by cell contact.
