促血管生成素-2在口腔鳞癌中表达的初步研究A Preliminary Study on the Expression of Angiopoietin-2 in Oral Squamous Cell Carcinoma

摘要(Abstract):
目的:研究口腔鳞癌组织中促血管生成素-2(Angiopoietin-2,Ang-2)的表达及其与临床病理学特征间的关系,初步探讨Ang-2在口腔鳞癌中的作用及意义。方法:免疫组织化学SABC法检测口腔鳞癌标本41例(有淋巴结转移者13例)及30例癌旁正常组织和10例正常口腔黏膜组织中的Ang-2表达。应用BioMias图像处理系统处理图像。SPSS11.0统计软件行数据处理。结果:Ang-2表达主要集中于肿瘤细胞胞浆,尤其是癌灶边缘的血管重塑区。Ang-2在口腔鳞癌组织中表达显著高于癌旁正常组织(P<0.01)及正常口腔黏膜组织中Ang-2的表达(P<0.01);Ang-2表达与肿瘤的淋巴结转移(P<0.01)密切相关,而与病人的性别、年龄和临床分期及肿瘤的分化程度无关(P>0.05)。结论:Ang-2在口腔鳞癌组织中的过度表达可能在口腔鳞癌的发生、发展中起重要作用。
Objective:To explore the role and the significance of angiopoietin-2 (Ang-2) expression and its relationship with clinicopathologic characteristics of oral squamous cell cacinoma (OSCC). Methods: Immunohistochemical staining (SABC) was used to quantify Ang-2 expression in 41 cases of OSCC(13 cases with lymph node metastasis), 30 cases of adjacent normal tissue and 10 cases of normal oral mucosa. The images were analyzed with BioMias system developed by Sichuan University. SPSS 11.0 software was used for statistical analysis. Results: Ang-2 was mainly detected in cytoplasm of OSCC, especially at sites of vascular remodeling in tumor margin. The expression of Ang-2 was significantly higher in OSCC than in adjacent normal tissues( P <0.01) and normal oral tissues( P <0.01).In respect of clinicopathologic factors, lymph node metastasis( P <0.01)was correlated with Ang-2 expression, but there was no sianificaut correlafion between the Ang-2 positive expression and patients’ sex,age, TNM stages or tumor’s differentiation degrees( P >0.05).Conclusion: Overexpression of Ang-2 in OSCC may play a potential role in the development and progression of OSCC.
