苗药杆努尽烟对慢性支气管炎大鼠肺组织TLR4及TNF-α表达的影响

摘要(Abstract):

目的:研究苗药杆努尽烟对慢性支气管炎模型大鼠肺组织Toll样受体4(TLR4)及肿瘤坏死因子-α(TNF-α)水平的影响。方法:50只SD大鼠分为正常对照组、慢性支气管炎模型组、杆努尽烟低剂量组(2 g/kg)、杆努尽烟高剂量组(8 g/kg)及桂龙咳喘宁阳性药物组,除正常对照组外,其余各组通过烟熏加脂多糖气管注射法建立慢性支气管炎大鼠模型;观察各组大鼠呼吸系统症状及肺组织病理变化,ELISA法检测各组大鼠肺组织肿瘤坏死因子-α(TNF-α)含量,Western Blot法及q PCR法分别检测各组大鼠肺组织TLR4蛋白及mRNA水平。结果:与模型组相比,高、低剂量杆努尽烟均可改善慢性支气管炎大鼠呼吸系统症状,减轻肺组织炎症,降低慢支大鼠肺组织中TNF-α的水平,减少TLR4蛋白及mRNA的表达(P <0. 05),其中高剂量杆努尽烟效果更好。结论:苗药杆努尽烟可减轻慢性支气管炎模型大鼠呼吸道症状及肺组织炎症,其机制可能与下调大鼠肺组织TLR4及TNF-α表达水平有关。
Objective: To study the effects of Lysionotus pauciflorus Maxim. on the levels of TLR4 and tumor necrosis factor-α( TNF-α) in lung tissue of chronic bronchitis rats. Methods: 50 SD rats were divided into normal control group,chronic bronchitis model group,low dose group( 2 g/kg),high dose group( 8 g/kg) and Guilong Kechuanning positive drug group. The rat model of chronic bronchitis was established by fumigation and endotracheal injection of lipopolysaccharide( LPS) in other groups except the normal control group. The respiratory symptoms and pathological changes of lung tissue were observed,and the content of tumor necrosis factor-α( TNF-α) in lung tissue of rats in each group was detected by ELISA method. The levels of TLR4 protein and mRNA in lung tissue of rats in each group were detected by Western Blot and qPCR. Results: Compared with the model group,the high and low dose of Lysionotus pauciflorus Maxim. could improve the symptoms of respiratory system,alleviate the inflammation of lung tissue,decrease the level of TNF-α and the expression of TLR4 protein and mRNA in the lung tissue of chronic bronchitis rats( P < 0. 05). Among them,the effect of high dose bar was better. Conclusion: Lysionotus pauciflorus Maxim. can alleviate the respiratory symptoms and inflammation of lung tissue in rats with chronic bronchitis. The mechanism may be related to the change of TLR4 and TNF-α expression in rat lung tissue.
