摘要(Abstract):
目的:考察过氧化氢对H9c2心肌细胞及原代心肌细胞损伤作用的相似性。方法:选用H9c2心肌细胞与新生大鼠原代心肌细胞,应用MTT法测定不同浓度过氧化氢对H9c2心肌细胞及原代心肌细胞活性的影响,观察两株细胞对过氧化氢损伤的敏感性。结果:600~1 600μmol/L的过氧化氢对H9c2心肌细胞及原代心肌细胞所造成的氧化损伤均具有浓度依赖性,1 200μmol/L的过氧化氢使H9c2心肌细胞的存活率降低40%~60%(P<0.01),表现出与原代心肌细胞相似的H2O2损伤敏感性(P>0.05)。结论:H9c2心肌细胞与原代心肌细胞对过氧化氢氧化损伤具有相似的作用和浓度依赖性。
Objective: To explore the similarity of the damage effect of hydrogen peroxide(H2O2) on cardiac cell H9c2 and primary cardiac myocytes.Method: H9c2 cells and primary cardiac myocytes of neonatal rats were selected,and the effects of different concentration of H2O2 on the activity of H9c2 cells and primary cardiac myocytes were measured with MTT method,and their sensitivity to oxidative damage induced by H2O2 was observed.Result: In the concentration range of 600~1 600 μm·L-1,H2O2 caused oxidative damage to these two kind of cells in a dose-dependent manner.The viability of H9c2 cells was reduced by 40%~60% when the concentration of H2O2 was 1 200 μm·L-1,which was similar to that of primary cardiac myocytes.Conclusion: H2O2 has similar oxidative damage effect on H9c2 cells and primary cardiac myocytes.
