β淀粉样肽对神经母细胞瘤细胞Notch信号通路的影响
Influence of β-amyloid Peptide on Notch Signal Transduction in Neuroblastoma Cells

摘要(Abstract):

目的:研究β淀粉样肽(Aβ)处理SH-SY5Y细胞以及α3神经型尼古丁受体(α3 nAChR)沉默细胞后细胞Notch信号通路的变化。方法:2μmol/L Aβ处理SH-SY5Y细胞以及α3 nAChR沉默细胞,用实时荧光定量聚合酶链式反应(Real Time PCR)和蛋白印迹(western blot)方法分别检测细胞中Notch信号通路中Notch1和Hes1的mRNA及蛋白水平的变化,评价Aβ对细胞Notch信号通路的影响。结果:Aβ处理的SH-SY5Y细胞以及α3 nAChR沉默细胞中Notch1和Hes1的mRNA及蛋白水平表达均增加,且在α3 nAChR沉默细胞中增加更为明显,差异有统计学意义(P<0.05)。结论:Aβ可增加细胞Notch信号通路基因表达,α3 nAChR沉默会增强Aβ对Notch信号通路基因表达的影响,这可能与阿尔茨海默氏病(AD)的发病有一定的关系。
Objective: To observe the changes of Notch signal transduction pathway in SH-SY5Y neuroblastoma cells and 3 nAChR silence cells after treated with β-amyloid peptide( Aβ). Methods: SH-SY5Y cells and 3 nAChR silence cells were exposed to 2 μmol / L Aβ1- 42. The mRNA and protein levels of Hes1 and Notch1 of Notch signal transduction pathway were measured by using Real-Time PCR and Western Blotting methods respectively. Results: mRNA and protein levels of Notch1 and Hes1 increased both in Aβ treated SH-SY5Y cells and 3 nAChR silence cells,and specially in 3 nAChR silence cells,the increase was more obvious. The differences were statistically significant( P < 0. 05). Conclusions: Aβ may enhance the gene expression in Notch signal transduction pathway; and the silence in α3 nAChR cells can enhance such effects,which might be connected to the pathogenesis of AD.
