人工繁育恒河猴CCR5全基因突变特征及其与人和部分非人灵长类动物的异同

摘要(Abstract):

目的:研究人工繁育恒河猴趋化因子受体5(CCR5)全基因突变特征及其与人和部分非人灵长类动物的异同。方法:对82只人工繁育恒河猴CCR5进行全基因测序,分析CCR5全基因序列中的突变位点及突变类型;使用DNASTAR、MEGA、Dna SP软件分析其与人及黑猩猩、猩猩、恒河猴、大猩猩和非洲绿猴其他非人灵长类动物的异同。结果:恒河猴群CCR5全基因序列中共存在8个突变位点,均为同义突变;恒河猴与人类CCR5基因DNA序列有31个位置上存在碱基差异,8个位置的编码氨基酸不同,3个突变位点与人CCR5基因突变情况相同;在与人类及非人灵长类动物中的CCR5基因序列比较中,DNA序列共有44个位点存在差异,氨基酸序列共有14个位点存在差异,非人灵长类动物CCR5基因均未发现Δ32缺失。结论:人工繁育恒河猴群CCR5基因突变具有与其他非人灵长类及人类不同的特异分布特征。
Objective: To study the mutation characteristics of CCR5 gene in artificial breeding rhesus monkey and its differences with human and some non-human primates. Methods: CCR5 gene was sequenced in 82 artificially bred rhesus monkeys,and the mutation sites and types of CCR5 gene were analyzed. The similarities and differences of human,chimpanzee,orangutan,rhesus monkey,gorilla,African green monkey and other non-human primates were analyzed by DNASTAR,MEGA and Dna SP software. Results: There were 8 mutation sites in the CCR5 gene sequence of the rhesus monkeys,all of which were synonymous mutations. In rhesus monkey and human CCR5 gene,there were 31 nucleotide differences in the DNA sequence,the coding amino acids in 8 sites ee different,and three mutation sites were the same as those in human CCR5 gene. In comparison with the CCR5 gene sequence of human and non-human primates,there were 44 different loci in DNA sequence and 14 in amino acid sequence,and no Δ32 deletion was found in CCR5 gene of non-human primates. Conclusions: The mutation of CCR5 gene in artificially bred monkeys has different specific distribution characteristics than other non-human primates and humans.
