miR-125a-5p对子宫内膜癌RL95-2细胞增殖凋亡的影响

摘要(Abstract):

目的:探讨miR-125a-5p对子宫内膜癌RL95-2细胞增殖、凋亡的影响及可能作用机制。方法:取对数生长期RL95-2细胞随机分为未转染组、mimic阴性对照组和mimic转染组,做相应处理后24 h,采用Real-time PCR检测miR-125a-5p表达,MTT检测细胞增殖,流式细胞术分析细胞凋亡,Western blot检测凋亡相关蛋白(Bcl-2、Bax、cleaved caspase3、cytochrome C)和信号通路相关蛋白(p-AKT、AKT、p-mTOR、m TOR)的表达。结果:同mimic阴性对照组比较,mimic转染组中miR-125a-5p表达显著增加,细胞增殖抑制率和细胞凋亡率明显升高,Bax、cleaved caspase3表达水平显著上调,Bcl-2和线粒体cytochrome C、p-AKT、p-mTOR的表达水平明显下降。结论:miR-125a-5p能够抑制子宫内膜癌RL95-2细胞增殖,机制可能与其改变PI3K/AKT/m TOR通路活性及调节凋亡相关基因有关。
Objective: To explore the effect and the mechanism of miR-125 a-5 p on proliferation and apoptosis in endometrial carcinoma RL95-2 cells,and to provide a new target for the clinical treatment of the disease. Methods: Human endometrial carcinoma RL95-2 cells in logarithmic growth period were randomly divided into three groups: untransfected control group,mimic negative control group,mimic transfected group. The Real-time PCR was used to detect expression of miR-125 a-5 p. MTT assay was used to assess cell proliferation. Flow cytometry was used to analyze cell apoptosis. Western blot was employed to test expressions of apoptotic proteins(bcl-2,Bax,cleaved caspase3,cytochrome C) and signaling pathway-related proteins(p-akt,AKT,p-mtor,m TOR). Results: The expression level of miR-125 a-5 p significantly increased in mimic transfected group compared with mimic negative control group. Similarly,the inhibitory rates of proliferation and apoptotic rates were increased,the expression of Bax and cleaved caspase3 were markedly up-regulated,and the expression of Bcl-2,mitochondria cytochrome C,p-AKT and p-mTOR were down-regulated. Conclusion: MiR-125 a-5 p can inhibit the proliferation of endometrial carcinoma RL95-2 cells,the mechanism may be related to its effects of changing PI3 K/AKT/m TOR pathway activity and regulating the apoptosis-related gene.
