氟化钠对小鼠肝脏细胞色素P4501A1酶活性的影响

摘要(Abstract):

目的:研究高氟对细胞色素P4501A1酶活性的影响,为探讨高氟地区肿瘤的发生发展提供实验依据。方法:昆明种雄性小鼠120只,分3个实验组和1个对照组,每组30只;实验组分别饮用50 mg/L、200 mg/L、400 mg/L NaF水溶液,于2周、5周、8周后,分别用氟离子选择电极法和分光光度法测定小鼠骨氟含量与肝微粒体中CYP1A1酶的活性。结果:实验组与对照组相比CYP1A1酶活性受到诱导,诱导率为2.1%~121%,但各剂量组之间无显著性差异,8周与2周相比酶活性显著增强。结论:NaF可不同程度地诱导CYP1A1的酶活性,摄入过量氟对肿瘤的发生发展具有一定的影响。
Objective: To investigate the effects of sodium fluoride(NaF) on the enzymic activities of cytochrome P4501A1 in mouse liver.Methods: One hundred and tweenty male mice were divided into 4 groups with 30 animals in each.Group 1 was negative control(animals of group 1 were fed with tap water),and the other 3 groups were fed with fluoridated drinking water in which NaF concentrations were 50 mg/L,200 mg/L,and 400 mg/L respectively.Fluorine content in bone and enzymic activities of CYP4501A1 in liver microsomes were detected by using fluoride ion selective electrode and spectrophotometry separately in 2,5,and 8 weeks.Results: Enzymic activities of CYP1A1 in 3 experimental groups were induced by NaF and the induced rates ranged from 2.1% to 121%,but there was no significant difference among the 3 groups.CYP1A1 activities at the 8th week were significantly higher than those at the 2rd and 5th weeks.Conclusion: NaF could induce enzymic activities of CYP1A1 suggesting that excessive fluorides might have some influence on pathogenesis and development of carcinoma.
