存放全血温度及时间对提取DNA质量的影响

摘要(Abstract):

采用PCR扩增评估提取DNA的质量 ,研究在不同温度、时间条件下存放全血对提取DNA的影响。结果 :不同温度 ( 4℃ 1~ 4d、- 2 0℃ 3个月、- 85℃ 3个月 )条件下存放的全血 ,4℃存放 1~ 4d与 - 85℃存放 3个月无显著差异 ,4℃存放 1~ 4d与 - 2 0℃存放 3个月、- 2 0℃与 - 85℃存放 3个月呈显著差异。结论 :4℃ 1~ 4d及 - 85℃ 3个月 ,能获得较高质量的DNA样品
A study of the methods of extracting DNA from whole blood stored in different temperature conditions. Using hypoosmotic hemolysis method and PRC method to evaluate the quality of the extracted DNA. DNA Samples extracted from the whole blood stored at temperature of 4℃ for 1 to 4 days,-20℃ and -85℃ and for 3 months at were compared. The result showed that no significant difference was found between 4℃ and -85℃,but there was a significant difference between -20℃ and -85℃. Samples of whole blood with comparatively high quality could be obtained when the whole blood was stored at the temperature of 4℃ for 1 to 4 days and at the temperature of -85℃ for 3 months.
