摘要(Abstract):
目的:研究灯盏细辛醇提物不同活性部位对β-淀粉样蛋白(Aβ)引起的神经母细胞瘤细胞(SH-SY5Y)凋亡的拮抗作用,探讨其是否在阿尔茨海默病(Alzheimer’s disease,AD)治疗中有神经保护作用。方法:选用灯盏细辛醇提物不同活性部位及Aβ25-35处理SH-SY5Y细胞,用比色法测定MTT还原率,Annexin V-FITC试剂盒测定细胞凋亡水平。结果:安全浓度的灯盏细辛醇提物不同活性部位能对抗Aβ引起的MTT还原率降低和细胞凋亡。结论:灯盏细辛醇提物不同活性部位可对抗Aβ的神经毒性作用。
Objective:To investigate the effect of active parts extracted with alcohol from Herba Erigerontis(HE) against apoptosis in neuroblastoma(SH-SY 5Y) cells induced by β-amyloid peptide(Aβ),and reveal whether the drug has an effect of neural protection in the therapeutic strategy of Alzheimer disease.Methods: SH-SY 5Y cells were treated with different active parts extracted from HE,and then exposed to Aβ25-35.MTT reduction was detected by using colorimetric method and apoptosis rates were analyzed with Annexin V-FITC reagent kit.Results: The active parts of HE at a safe concentration inhibited MTT reduction and decreased apoptosis rate resulted from Aβ.Conclusion: The active parts from HE may be important and can play an effect against neurotoxicity of Aβ.
