阿尔茨海默病患者大脑生物膜性抗氧化物质含量的改变

摘要(Abstract):

选用阿尔茨海默病(AD)患者不同部位的尸解脑组织标本,以同龄非神经系统疾病患者尸解脑组织标本作为对照,测定脑组织中蛋白质、DNA、辅酶Q、缩醛磷脂酰乙醇胺和丙二醛含量,研究内源性生物膜性抗氧化物质在AD发生中的变化。结果显示:蛋白质和DNA含量两组间无明显差别;AD患者大脑皮质额叶和海马区域中辅酶Q含量升高,缩醛磷脂酰乙醇胺含量降低,经诱导后的丙二醛含量明显高于对照组。表明:AD患者受影响的脑组织中生物膜性抗氧化物质水平发生异常改变,这种表现可能与体内自由基含量的异常变化有关,本研究从另一方面反映了自由基在AD发生机制中的重要作用
For investigating the effect of biological membrane antioxidants in Alzheimer's disease(AD),the frontal and occipital cortexes, and hippocampus from human brains affected by AD were analyzed for their contents of protein, DNA, ubiquinone, phosphatidylethanolamine plasmalogen and malondiadehyde. The age matched control brains with same areas as AD were chosen from the patients died from causes unrelated to the nervous system. The results showed that there were no differences of protein and DNA amounts between the brains with AD and the controls; increased content of ubiquinone and declined level of phosphatidylethanolamine plasmalogen were observed in the frontal cortex and hippocampus in AD brains . Raised amount of malondiadehyde was present after being induced by iron in the brain areas affected by AD.No changes, however, appeared in the occipital cortex of brains from the patients with AD. These results demonstrate that levels of biological membrane antioxidants are modified in brains affected by AD, which is related to the change of free radicals. This study gives out one evidence in which free radials and the resulting toxicity might have an important role in pathogenesis of AD.
