高效液相色谱法测定紫珠果中齐墩果酸和熊果酸
Determination of Oleanolic Acid and Ursolic Acid in Zizhu Fruit by High Performance Liquid Chromatography

摘要(Abstract):

目的:建立紫珠果药材中齐墩果酸和熊果酸的含量测定方法。方法:采用RP-HPLC法,Phenomenex Kromasil ODS-Cl8(4.6 mm×250 mm,5μm)色谱柱;流动相为甲醇-水(90∶10);流速1.0 ml/min;柱温25℃;检测波长为215 nm。结果:齐墩果酸在0.011 5~0.103 5 g/L呈良好的线性关系;熊果酸在0.030 32~0.272 88g/L呈良好的线性关系;齐墩果酸平均回收率为99.70%,RSD为0.32%(n=9),熊果酸平均回收率为100.06%,RSD为0.87%(n=9)。结论:该方法操作简便快速,结果可靠,重复性好,可为有效控制紫珠果药材及其制剂的质量提供依据。
Objective: To establish a high performance liquid chromatography(HPLC) method for determination of oleanolic acid and ursolic acid in Zizhu fruit.Methods: The HPLC method was carried on C18(4.6 mm×250 mm,5 μm)column using methanol-water(90∶10,V:V) as the mobile phase,and detection wavelength was 215 nm.The flow rate was 1.0 ml/min and the temperature of column 25 ℃.Results: The calibration curve for oleanolic acid was linear in the range of 0.011 5~0.103 5 g/L,and the calibration curve for ursolic acid was linear in the range of 0.030 32~0.272 88 g/L,The average recovery of oleanolic acid was 99.70%,and RSD was 0.32%(n=9),while average recovery of ursolic acid was 100.06%,RSD 0.87%(n=9).Conclusions: The method is convenient,precise,reliable and with good reproducibility for determining the contents of oleanolic acid and ursolic acid in Zizhu,which may provide basis for effective quality control of Zizhu fruit products.
