灯盏乙素对β淀粉样蛋白诱导的神经母细胞瘤细胞中NMDAR1表达的影响

摘要(Abstract):

目的 观察灯盏乙素(Scu)对β淀粉样蛋白(Aβ1-42)诱导的SH-SY5Y细胞模型中N-甲基-D-天门冬氨酸受体亚型1(NMDAR1)的影响。方法 将神经母细胞瘤(SH-SY5Y)细胞分为对照组、Aβ寡聚体处理组、Scu处理组和Scu+Aβ寡聚体处理组,采用蛋白印迹法检测各组细胞中NMDAR1的蛋白表达情况,实时荧光定量PCR检测NMDAR1 mRNA表达水平,荧光探针法测定各组细胞氧自由基(ROS)的水平,黄嘌呤氧化酶法测定各组细胞超氧化物歧化酶(T-SOD)的活力。结果 与对照组比较,Scu处理组NMDAR1蛋白、NMDAR1 mRNA表达、ROS含量及T-SOD含量比较,差异无统计学意义(P> 0.05);Aβ寡聚体处理组细胞NMDAR1蛋白和NMDAR1 mRNA表达水平升高、细胞ROS含量升高、细胞T-SOD含量降低,差异均有统计学意义(P<0.05);与Aβ寡聚体处理组比较,Scu+Aβ寡聚体处理组NMDAR1蛋白和NMDAR1 mRNA表达水平下调、细胞ROS含量降低、细胞T-SOD含量升高,差异均有统计学意义(P<0.05)。结论 Scu可能通过下调NMDAR1 mRNA、减少NMDAR1蛋白表达,从而对抗Aβ寡聚体的毒性。
Objective To observe the effect of scutellarin(Scu) on N-methyl-D-aspartate receptor subtype 1( NMDAR1) in the cell model induced by β-amyloid protein( Aβ1-42).Methods Neuroblastoma(SH-SY5 Y) cells were divided into control group, Aβ oligomer treatment group, Scu treatment group and Scu + Aβ oligomer treatment group. Protein expression of NMDAR1 was detected by protein printing, and the expression level ofNMDAR1 mRNA was detected by real-time fluorescence quantitative PCR. The levels of cell oxygen radicals( ROS) were determined by fluorescence probe method, and the activity of cell superoxide disoxidase(T-SOD) was determined by xanthine oxidase method.Results Compared with the control group, NMDAR1 protein, mRNA expression, and oxidative stress index in Scu treatment group were compared, and the difference was not statistically significant(P> 0. 05). Cell NMDAR1 protein, mRNA expression levels, and cell ROS content were increased in Aβ oligomer treatment group. Cell T-SOD content was decreased, and the differences were statistically significant(P< 0. 05). Compared with the Aβ oligomer treatment group, the NMDAR1 protein and mRNA expression levels were lowered in the Scu + Aβ oligomer treatment group,and the cell ROS content decreased. The cell T-SOD content increased, and the differences were statistically significant(P< 0. 05).Conclusion Scu may combat the toxicity of Aβ oligomers by downgradingNMDAR1 mRNA and reducing NMDAR1 protein expression.
