下调干扰素诱导蛋白16对人主动脉外膜成纤维细胞凋亡的影响
Downregulating Effect of IFI16 Expression Downregulating on Human Aortic Adventitial Fibroblasts Apoptosis

摘要(Abstract):

目的:探讨下调干扰素诱导蛋白16(IFI16)对人主动脉外膜成纤维细胞(HAAFs)凋亡的影响及机制。方法:在HAAFs中转染IFI16小干扰RNA(siRNA)和Control siRNA,分别作为IFI16基因沉默组(Ⅰ组)、阴性对照组(C组),未经任何干预的HAAFs作为空白组(N组),用流式细胞仪检测凋亡,Western blot检测IFI16、活化凋亡蛋白聚腺苷二磷酸核糖聚合酶(Cleaved PARP)蛋白表达。结果:与C组和N组相比,Ⅰ组中IFI16蛋白表达减少,细胞凋亡率下降(P<0.05),伴随着Cleaved PARP蛋白表达减少。结论:下调IFI16表达可抑制HAAFs细胞凋亡,其机制可能与下调Cleaved PARP蛋白有关。
Objective: To investigate the effect and mechanism of silencing interferon-inducible protein 16( IFI16) on the apoptosis of human aortic adventitial fibroblasts( HAAFs). Methods: The specific small interference RNA( siRNA) and Control siRNA of IFI16 were transfected into HAAFs in vitro instantaneously. HAAFs were divided into three groups,IFI16 siRNA transfection group( Ⅰgroup),negative control group( C group),untreated HAAFs as blank group( N group). Then cell apoptosis was analyzed by flow cytometry. The protein levels of IFI16,Cleaved PARP were measured by Western Blot. Results: Comparing with group C and N,the protein expression levels and IFI16 were decreased in group I,the cell apoptosis was downregulated( P < 0. 05),accompanied with decrease of protein expression of Cleaved PARP. Conclusion: Downregulating of IFI16 expression can inhibit HAAFs cell apoptosis,which may be partially related to the downregulating of Cleaved PARP expression.
